Compound Of Artemisia Annua Oil Quality Standards Study

The formula of Compound Artemisia oil ointment is composed of Artemisia oil, RadixSopborae Flavescentis, Radix Stemonae, Flos Lonicera Japonicae, and Menthol, with thefunction of Heat-clearing and Detoxifying, dispelling wind and dampness, destroying parasitesand relieving itching. It is used to cure itch dermatoses belonged to the wind-heat type ordamp-heat type. To evaluate the quality of the preparation, qualitative and quantitative methodsfor the quality control of compound Artemisia oil ointment was established, and the preliminarystability was inspected. Five kind of medicinal materials in the preparation was identified byTLC, while Camphor and Eucalyptol , Matrine and Oxymatrine, Radix Stemonae, Chlorogenicacid, and Menthol were set as reference substance. The samples displayed the spots of the samecolour as the reference substance or control medical materials did in the corresponding places ofthe Chromatography, while the negative sample did not. The method is rapid, simple, specific ,and the spots were fairly clear. Taking Artemisia ketone and Matrine as indexes, RP-HPLCmethods were developed to determine Artemisia oil and Radix Sopborae Flavescentis in thepreparation. The external standard method was used to calculate contents with area of peak. Thechromatographic conditions were as follows: A Kromasil C₁₈ column (250 mm×4.6 mm, 5μm)was used. For Artemisia ketone, mobile phase consisting of methanol-water (80∶20) wasadopted, and the detecting wavelength was set at 243 nm. The column temperature was 30℃.The flow rate was 0.8 mL·min^-1, and the sample size was 10μL. For Matrine, mobile phaseconsisting of acetonitrile-0.1%phosphate(pH adjusted to 7.0 with Triethylamine) (20∶80) wasadopted, and the detecting wavelength was set at 205 nm. The column temperature was 30℃.The flow rate was 1.0 mL·min^-1 , and the sample size was 10μL. The calibration curve ofArtemisia ketone Matrine showed good linearity over the range of 0.22045～3.5272μg, and theregression equation is A=104.6099C+0.8614 with r= 1.0000. The average recovery was 98.37%with RSD 0.34%. Meanwhile, the calibration curve of Matrine showed good linearity over therange of 0.505～4.040μg ,and the regression equation is A=41.4601C-0.8001with r=0.9999.The average recovery was 95.78 % with RSD 1.01%. The method is sensitive, accurate andrapid. The results showed that the established standard are suitable for the quality control ofCompound Artemisia oil ointment , and the preparation is of stable quality at room temperaturewithin 3 months.
