| Objective To reveal the influence of electro-acupuncture therapy on mitochondria ultrastructure,the cytochrome C oxidase activities and the glucose regulated protein 75 of hippocampus of SAMP8 and partly explore the mechanism of electro-acupuncture therapy on Alzheimer's Disease.Methods SAMP8 were punctured on "Baihui(Du20)" and "Yongquan(K1)" according to the therapeutic principle of "Tonifying kidney and activating Du meridian to reviving brain and promoting intelligence".The treatment was applied once daily and totally lasted for 21 days.The changes of learning and memory abilities were measured by Morris water maze test.The bulk density and area density were detected by transmission electron microscope(TEM).The activities of cytochrome c oxidase(COX) in hippocampal mitochoneria were detected by spectrophotometric method and the expressions of grp75 were detected by immunohistochemistry method.Results 1,Comparing with control group,the average escape latency of electro-acupuncture group was decreased(P＜0.05);Comparing with control group(30.27±6.12s),the times of swim in primary platform quadrant of electro-acupuncture group(39.55±5.47s) were increased (P＜0.05).2,Comparing with control group(0.354±0.010),the bulk density of electro-acupuncture group(0.601±0.025) are increased (P＜0.01 );Comparing with control group(1.167±0.051 ),the area density of electro-acupuncture group(1.291±0.071) are increased (P＜0.05).3,COX activity of electro-acupuncture group(0.016±0.003) is higher than control group(0.011±0.004)(P＜0.01).4,Comparing with control group(23.54±2.71),the expressions of grp75 of electro-acupuncture group(29.53±4.16)are increased(P＜0.05).Conclusion The curative mechanism of electro-acupuncture may be related to protect mitochondrial structure and function and improve energy metabolism by relieving the damage of mitochondrial ultrastructure,increasing activity of COX and enhancing the expressions of grp75. |
