IntruduceAcute myocardial infarction (AMI) is a serious global threat to human health. Acute myocardial infarction incidence rate of 45 to 55/10 million people in china,with thrombolytic therapy and angioplasty,such as acute reperfusion therapy and drug treatment application, AMI treatment has been achieved a great progress in the past 20 years. But were able to simply resume their death myocardial infarction area of activity infarct zone with the contraction due to cardiac cells to reduce the amount of scar tissue to replace, myocardial fibrosis(MF),ventricular remodeling and ultimately to the development of heart failure. Recently,as stem cell research has been constantly deepened, different sources of stem cell can be vitro isolation,culture,amplification using biological technology,it's expected that function of myocardial remodeling treatment(CCM) as a treatment for heart disease and the new model may become the treatment of myocardial infarction shortcut,show that good clinical application. However,the current stem cell treatment of AMI still exist many problem,which means the choice of transplantation has been more and more attention.ObjectiveBy the following aspects of the study: 1: Point-injection of mesenchymal stem cells on the impact of the infarcted myocardium:(1) observe whether there is a 5-bromo deoxy nucleoside uridine (Brdu) labeled cells;(2) Brdu is What kind of cells; 2:further explore Mesenchymal stem cells (MSCs) point injection of acute myocardial infarction after infarction and the surrounding area early remodeling of collagen.Evaluate the point injection MSCs transplantation as a new way of the feasibility and efficacy of treatment for AMI, prevention and treatment of myocardial necrosis,collagen remodeling to provide new, safe,effective and feasible means.MethodsThis study though MSCs isolated,culture,amplified after acute myocardial infarction corresponding point injection,to explore new points of MSCs transplantation as a way to transplant the feasibility of observation points after transplantation in myocardial infarct- -ion and MSCs distinguish between the junction of the situation and the impact of collagen remodeling. Through the left ventricle coron- -ary artery ligation rabbit (LVB),the establishment of AMI model, the models were injected MSCs in four points after seven days, infarcte after the first five weeks will be put to death in rabbits.ResultsPoint-injection stem cell were found Brdu the MSCs in myoca- -rdial infarction zone and zones at the junction,Brdu positive staining the cells are also cardiac-specific troponin T(cTnT) posit- -ive,the migration of these cells have to differentiate into cardiac cells.Masson staining showed that stem cell transplantation group and the model and point injection saline group:infarction in the central area at the junction of the region and integration of less collagen fibers,are more orderly,and the central area of infarction and the region at the junction of collagen fibers Ban Kuaizhuang integration obvious,With disorder.Each group collagen volume fra- -ction (CVF) no significant difference (p>0.05) but the normal group, point injection stem cells,point injection saline mix model to compare CVF was followed by increase in myocardial infarction after 7 Heavenly stems cell transplanted stem cells before the point injection group,point injection saline serum matrix metalloprotei- -nase enzyme-1 (MMP-1)levels showed no significant difference (p>0.05),myocardial infarction after seven days the two groups of serum Organization of matrix metalloproteinase enzyme inhibitor -1 (TIMP-1) levels showed no significant difference (p>0.05), Point infarction after five weeks ago and the injection of stem cells injected with normal saline points respectively compared serum MMP-1,TIMP-1 levels were significantly different (p<0.05).Conclusion1. acupoint injection therapy of bone marrow mesenchymal stem cell transplantation in the treatment of myocardial infarction is safe, effective and feasible,which can significantly improve myocardial collagen remodeling after myocardial infarction.2. allogeneic bone marrow-dirived mesenchymal stem cell transplantation for myocardial infarction impact:(1) reduce the collagen remodeling of a control factor in the MMP-1 levels in the serum.(2)reduce the non-infarcted area collagen deposition,slow infarct zone collagen deposition,inhibition of myocardial fibrosis and repair of myocardial fibrosis, reversing myocardial collagen network reconfiguration.(3) to prevent the integration of collagen fibers,the protection of the basic structure of myocardial tissue orderly and reduce ventricular remodeling .This may be antologus bone marrow-derived mesenchymal stem cell transplantation to improve left ventricular function after myocardial infarction in one of the mechanisms.3. Emerging as a method of treatment AMI,MSCs Point allogeneic transplantation The method is simple,economical and effective, and progress is expected to improve the prognosis of patients with AMI,for the treatment of cardiovascular disease provide a good source of seed cells,ideas and have good clinical application.
