Depression(抑郁症) is a group of mood disorders(心境障碍) or affective disorders(情感性障碍),which caused by a variety of reasons as the main symptoms of depression,is a group of self-experience of mood in depression as the central clinical symptoms or state.In the mental diseases,the incidence of depression second only to the schizophrenia,and about 15%of patients with depression with suicidal tendencies,which is not conducive to social stability and economic development.Depression because of its high incidence and difficult to cure,serious side effects of traditional antidepressant drugs acting on the single aspect or single-target of the disease,the relatively high cost,easily recurrence and so on,so the research and development of new,safe and effective antidepressant drugs will be very benefit to social and economic.Liquiritint is extract from Ganmai Dazao Decoction,a traditional Chinese medicine prescription of depression.The result of demolition side comparison test,as well as the single drug experiments,the effective separation of parts,combined with animal experiments,indicates that liquiritint as the major components of antidepressants.On the basis of the result,we begin to explore the pharmacological effects and the mechanism of liquiritin.1 Study on the protective effect of nerve cells in vitro of liquiritin1 The protective effect on BV2 ceils of liquiritinIn the 96-well plates of normal growth BV2 cells,respectively,plus different doses of liquiritin solution,2 hours later,the glutamic acid solution is added to establish BV2 cells glutamate injury model,40 hours later,MTT cell viability assay.The same method,in 6-well plates of normal growth of BV2 cells is added a certain amount of liquiritin solution,2 hours later the glutamic acid solution is added to establish BV2 cells glutamate injury model,40 hours later,Annexin V - FITC kit is used to detect apoptosis. The experimental results show that liquiritin can not fully prevent the damage caused by glutamate BV2 cell death,but can reduce their mortality; and liquiritin delay to some extent,glutamate injury BV2 cell apoptosis.2 liquiritin -containing serum on the protective effects of nerve cellsMethods in accordance with the above glutamate injury model of BV2 cells, respectively,by adding a blank serum,doses of glycyrrhizin-containing serum,MTT cell viability assay results showed that the cell protective effect was not obvious,but the role of cell morphology and trends,or have a certain influence,its role has yet to be confirmed.2 Antidepressant mechanism on nerve receptors of liquiritin1 Influence on CUMS rats sucrose preference of liquiritinEstablish animal model of chronic stress of rats,feed liquiritin according to designed doses,carry out sugar test at the same time,record of sugar consumption,water consumption,calculated sucrose preference rate. Monitor the sucrose preference rate of CUMS rats before and after feed liquiritin,results show that liquiritin can be reversed the decline of CUMS rats with sucrose preference rate to some extent,reflects the role of antidepressants.2 effects on weight of CUMS rats of the liquiritinEstablish animal model of chronic stress of rat,according to liquiritin dose given at the same time record body weight.Compared body weight of CUMS rats before and after feed liquiritin showed that liquiritin has no significant effect on body weight in rats.May be related to the use of a relatively minor stimulus methods.3 effects on CUMS rat brain 5-HT1A receptor of liquiritinUse the immunohistochemical staining technology,dyeing the 5-HT1A receptor in the brain,using Image Pro Plus 6.0 image software to determine and analysis the cumulative optical density(IOD) of rat brain CA1 area,CA3 area of hippocampus,cingulate gyrus and hypothalamus,four brain regions, to compare the brain 5-HT1A receptor of each group animals.The results showed that chronic stress,the regions of the rat brain 5-HT1A receptor in a downward adjustment,after feeding fluoxetine as positive drug can reverse the above phenomenon;liquiritin given group on the various brain regions (except the cingulate gyrus) of the reduced 5-HT1A receptors also show a certain degree of role reversal,but the statistical tests there was no significant difference in each group.4 effects on CUMS rats brain 5-HT2A receptor of liquiritinDetermined using the same method dyeing rat brain hippocampal CA1 area, CA3 area of hippocampus,cingulate gyrus and hypothalamic accumulation of the four brain regions of optical density(IOD),to compare the brain 5-HT2A receptor density of animals in each group.The results showed that after chronic stress,the brain regions of the rat brain 5-HT2A receptor increases occur;give positive drug fluoxetine can reverse the above phenomenon;the area of the brain 5-HT2A receptor of liquiritin groups showed no effect increase in the reversal.In summary,glycyrrhizin and its drug-containing serum in vitro BV2 microglial cells of glutamate have a protective effect,indicating that the antidepressant activity of glycyrrhizin may be related to the protective effects of nerve cells,but as a result of liquiritin study in vivo metabolism of the lack of reliable information,this role has yet to be explored in depth; Animal experiments have shown that glycyrrhizin can significantly reverse the chronic mild stress model in rats to reduce the amount of sucrose preference,to ease the depression center both symptoms of excitement missing, glycyrrhizin have the exact effect to the model;liquiritin on the model of rat brain 5-HT receptor requires further study.
