The ingredients of traditional Chinese medicine are extremely complex.Before conducting animal experiments,we do not know ifa single component isolated is effective.But using the experiment in vitro,we can judge whether the ingredients in the body can be through the biofilm to be absorbed,and then we can select the effective material more quickly and effectively.The issue is one part of the national "973" program subject(the No.of the subject is 2007CB512605).In early experiments,we found that it is very important to choose an active ingredients of Chinese medicine absorbed well and easy to detect before analysing drugs in the body.For example,in the analysis of experiments done in vivo Huangbai we choose berberine as the active ingredients,but the oral absorption of berberine is very poor.Another example is that we chose Curculigoside as an active ingredient in vivo analysis of Curculigo,but we failed to detected Curculigoside in the blood.The reason for the failure may be that Curculigoside metabolisms in the body is metabolized very quickly,or changes to other substances very quickly.Thus,for this topic,forecasting for the absorption of Chinese medicines in vitro and combining the study in vivo is meaningful.The biopartitioning micellar chromatography(BMC) compared to other detection methods in vitro such as n-octanol/water system,lipid membrane chromatography,liposome/water system etc,is rapider,simpler,more economic and more reproducible.At this stage,as an economical method in vitro study of drug membrane permeability,BMC has been studyed comprehensively in the chemical drugs,and has great significance in guiding the drug design and the development of new drugs.In BMC,the unique amphiphilic of micellar in RP-HPLC provides hydrophobic,hydrophilic,hydrogen bond,space and electrostatic force between the molecules like biofilm.Drugs' retention behavior in the stationary phase to some extent reflected the transfer process in the biofilm. In the field of traditional Chinese medicine,the prediction of ingredients' oral absorption by the method of BMC has not yet been reported.So we used BMC with k as the standard to predict the oral absorption of 37 kinds of active ingredients of Chinese medicine.These active ingredients include berberine,aconitine,cinnamic acid,orcinol glucoside,etc.According to the literature reported and the result of our research,we think that using the BMC to predict the oral absorption is reliable in some extend,and can be the guideline.At the same time, combineing Aconite Decoction's fingerprint,we initially explored the oral composition which may be absorbed in Aconite Decoction.
