Objective:Observe the influence of JAK-STAT and MAPK pathway's signal transduction in RA synovial cell of experimental rabbits by moxibustion,and reveal the molecular signal mechanism of experimental RA by moxibustion.Methods:Use 30 Japanese white rabbits,which divided into three group—normal group,model group and moxibustion group by weight,sex randomly.Every group has 10 rabbits.Just injectd the Freund's Complete Adjuvant(FCA) into the joint cavity of two hind legs of model group and moxibustion group by 0.5ml/Kg.Therapy:Moxibustion on two ShenShu points 5 zhuang once a day and 6 days was one treatment course.There were totally 3 treatment course.Fix the normal group and model group by the same method but don't moxibustion.We use ruler to survey the perimeter of hind legs of rabbits before therapy and the 3th,6th,9th,12th,15th,18th,21th day of therapy.After the treatment of 21th day,take the knee joint synovial membrane organization.The author's research,by taking the synovium cell,which is a key tache in the pathology of RA and an important target of moxibustion as a breakthrough point,Apply gene chip and bioinformation analytical technique to detect the 96 sorts of signal molecule of JAK-STAT and MAPK pathway.Results:before the therapy,the perimeter of left and right knee joint of each group's rabbit have non-significance difference(P>0.05); After make the model,compared model group's perimeter of left and right knee joint to normal group,the data obviously elevated(P<0.01),which suggests that the inflammation model exists continually:After the treatment,the perimeter of moxibustion group's left and right knee joint compares the model group to drop(P<0.05),which prompts that Moxibustion have the anti-inflammation and detumescence function. Compare model group with normal group,JAK-STAT pathway has been activated abnormally,and as a result,the expression of signal molecule such as JAK3,STAT3,C/EBP beta,INDO has been up-regulated,while down-regulation of A2M,MIG,IL-2Rr;compare moxibustion group with model group,the expression of signal molecule such as JAK3,STAT3,C/EBP beta,INDO has been down-regulated,while up-regulation of IL22R.Conclusions:Moxibustion plays a very important roll of transparent inhibition in abnormal activation of JAK-STAT pathway's signal transduction in RA synovial cell of experimental rabbits.
