Objective Investigate influences of the serum containning Zuogui Pill(ZGP) and Yougui Pill(YGP) of mites on activity of aromatase from glioma cell.Method 1.Preparation of serum:312 female sexual matured mice were divided into saline group(13 period group with 6 mice in each group),ZGP group(13 period group with 9 mice in each group) and YGP group(13 period group with 9 mice in each group).Randomized grouping,corresponding medicines were fed once.They were fed on corresponding saline and medicines respectively.Dosage of ZGP group was 0.06/10g and YGP group 0.09g/10g.And every mouse was given as 0.3ml/10g. Sera were obtained 30min,1h,2h,3h,4h,5h,6h,7h,8h,10h,12h,18h,24h after medication.Separating serum,taking the serum 200μL above,adding distilled water 100μL.And the serum were modified in boiling water bath after Oscillation. Tking the upper layer medium by centrifuge(12000rpm,15min) for using.2.The analysis of blood concentration:Samples were analyzed by high perfor -mance liquid chromatography,make the qualitative judgment by comparing the different retention time and.peak shape among all samples.Selecting the most prominent change peak shape to analyse quantitatively,and draw the drug concentration-time curve.The medicated serum obtained in pre and post peak time was prepared for using.3.Culture cells:The C6 glioma cells had been maintained by normal method of stable growth.They were planted on 24 and 96 well culture plate.C6 glioma cells cultured with medicated serum at all time points and proper Te for 24h in 24 well culture plate,then indicate the mRNA expression of aromatase.We measured E₂ in the upper layer medium and examined cell proliferation after C6 glioma cells cultured for 48h in 96 well culture plate4.The recipe separated of ZGP and YGP according to prescription compo -sition,the same as above,we do the other experiment with the recipe.Results 1.The analysis of blood concentration:The high content of dispersion serum are ZGP and YGP 5h,6h,7h groups,decomposed recipe 3 15min group and decomposed recipes 30min,1h,4h groups.2.E₂ in the upper layer medium(pg/mL):E₂ in the upper layer medium in ZGP 7h group(241.99±24.14) of mice,YGP 5h group(241.47±16.61) of mice and YGP 6h group(379.47±115.05) of mice were higher than that saline group with Te (195.43±17.11).E₂ in the upper layer medium in decomposed recipe 3 30min group (132.35±14.06)of mice,decomposed recipe 3 15min group(135.15±12.36) of mice were higher than that saline group with Te(195.43±17.11).3.Cell proliferation:There were no significant proliferative changes in cell proliferation among ZGP group,YGP group,the recipe separated and saline group with Te.4.The mRNA expression of aromatase:There were no significant proliferative changes in the mRNA expression of aromatase among ZGP group,YGP group and saline group with Te.Conclusion ZGP and YGP can promote estrogen synthesis from glioma cell,this effect was independent with glioma Cell proliferation.And ZGP and YGP did not effect the mRNA expression of aromatase.
