Objective:To continue our prophase-study,we useed agonist and antagon of 5-HT1A receptor to affect electro-acupuncture and researched the changes of behavior and 5-HT expression.So as to provide study direction of exploring the central 5-HT1AR mechanism of anti-anxiety effect after morphine with- drawal by electro-acupuncture.Then provides experimental foundation to resist relapse after drug addition withdrawal by acupuncture on clinic.Methods:Establishd morphine withdrawal model first.1,The mice were randomly individed into model group,model and electro-acupuncture group,model and excitomotory group,model and excitomotory and eletro-acupuncture group,control group.Then the mice of the model and excitomotory group,model and excitomotory and eletro-acupuncture group were injected 8-OH-DPAT(0.2mg/kg) into abdominal cavity as 5-HT1A receptor excitomotory.2,The mice were randomly individed into model group,model and electro-acupuncture group,model and antagon group,model and antagon and eletro-acupuncture group,control group.Then the mice of the model and antagon group,model and antagon and eletro-acupuncture group were injected WAY100635(0.5mg/kg) into abdominal cavity as 5-HT1A receptor antagonist.The two acupoints "Baihui(DU20) and Sanyinjiao(SP6)" were selected to participate the electro-acupuncture with the stimulating power which kept the back legs of mice slightly shaken for 15min(thin-dense alternative wave,frequency 15-25/min),once a day,lasted for 3 days.The behavioral tests were measured by the elevated plus-maze test(EPM),immunohistochemistry with image analysis technique were used to investigate the effect of electro-acupuncture on the change of the optical density(OD) value and grey value of nuclei of median raphe,prefrontal cortex's 5-HT expressions in anxiety model after morphine withdrawal and the influence of 5-HT1Areceptor agonist and receptor antagonist. entries(OE%)were lower(P<0.01).(2) Comparing with model group,model and eletro-acupuncture group,model and excitomotory group,model and excitomotory and eletro-acupuncture group,model and antagon and eletro-acupuncture group's open-arms time(OT%)and the open-arms entries(OE%) were higher(P<0.05,P<0.01). And the model and eletro-acupuncture group,model and antagon and eletro-acupuncture group got close to the control group,meanwhile the model and excitomotory group were lower(P<0.05),and the model and excitomotory and eletro-acupuncture group were higher than the control group(P<0.05).2.5-HT express.(1) Compared with control group:the model group's optical density(OD) value of 5-HT in nuclei of median raphe and prefrontal cortex were lower(P<0.01), and grey value were higher(P<0.05);The model and antagon group's optical density(OD) value of 5-HT in nuclei of median raphe were lower(P<0.01),and grey value were higher(P<0.05);The model and antagon group were not difference with the model group.(2) Compared with model group:model and eletro-acupuncture group, the model and excitomotory group,the model and excitomotory and eletro-acupuncture group's optical density(OD) value of 5-HT in nuclei of median raphe and prefrontal cortex were higher(P<0.05,P<0.01),and grey value were lower(P<0.05).Meanwhile,there was no noticeable difference between these groups and control group.(3)Compared with model and antagon group,model and antagon and eletro-acupuncture group's grey value of 5-HT in nuclei of median raphe and prefrontal cortex were lower(P<0.05,P<0.01),and close to the normal.Conclusion:1.Anxiety effect after morphine withdrawal could be resisted by electro-acupuncture,and to coordinate with the 5-HT1A receptor agonist.So we think the 5-HT1A receptor is the important pathway to against anxiety with eletroacupuncture. 2.The effect ofeletro-acupuncture antianxiety aren't blocked by the 5-HT1A receptor antagonist.So it's possible to exist another pathway correlated with the effect of eletro-acupuncture antianxiety.
