Chuanxiong Blood Circulation Effective Parts Of A Preliminary Study

Chuanxiong is the dried rhizome of Umbelliferae plant Ligusticum Chuanxiong Hort.,which is being recorded in China pharmacopoeia 2005.It is usually used to activate blood circulation to dissipate blood stasis.It is pungent in taste,at the same time slightly warm in nature,and attributive to the liver,thole and arcula cordis merideans.The efficacies include promoting blood flow,wenting,eliminating wind and alleviating pain.Chuanxiong has the certain efficacies of activating blood circulation and dissipating blood stasis;the doctors in past dynasties used Chuanxiong for activating blood circulation and dissipating blood stasis;the circs of clinical applications and modern study of pharmacology suggest that Chuanxiong can assuredly activate blood circulation and dissipate blood stasis,it can clinic disease of heart,brain,blood vessel and gynecological;so it possesses well foreground of exploitation.There are some literature information that Chuanxiong's chloroform fraction possesses the efficacies of activating blood circulation and dissipating blood stasis. The chloroform fraction was separated by silica gel chromatography,then isolated and purified by silica gel and reverse silica gel chromatography,six compounds were isolated and identified as sinapic acid,β-sitosterol,Z-6,8',7,3'-diligustilide,ferulic acid,chuanxingol,pregnenolone by UV,MS,¹H-NMR,¹³C-NMR methods.Two compounds which sinapic acid,pregnenolone are firstly separated from Ligusticum Chuanxiong Hort.Use methods of thin layer chromatography to appreciate the pregnenolone in ten batches of chloroform extractive;and established a HPLC method for determination of the pregnenolone in effective fraction and Chuanxiong,and determined the content in ten batches of effective fraction and Chuanxiong,we suggest that the content of pregnenolone should be more than 6.8%in effective fraction,and should be more than 0.32%in Chuanxiong,the method is sensitive,accurate and stable.This method can use to control the quality of chloroform extractive. Studied on the fingerprint of the effective fraction by HPLC,established the HPLC-FPC of effective fraction,we suggest that similarity of the effective fraction compared with HPLC-FPC should be more than 0.9,this method using for controlling the quality of the effective fraction is simple and have great stability and precision as well as repeatability,and it can be applicable broad.From the research,we separated six compounds from the effective fraction,and established a HPLC method for determination of the pregnenolone in effective fraction from Chuanxiong,and established the HPLC-FPC of effective fraction.This established the found for Chuanxiong' exploitation and application.
