Objective:To investigate the relationship between low-grade inflammation,oxidative stress and adiponectin in T2DM and early diabetic nephropathy.Methods:Sixty patients with T2DM(including 30 patients with early diabetic nephropathy)and 30 healthy controls were randomly collected.Serum 8-isoprostaglandinF2αand adiponectin were detected by an enzyme immunoassay(EIA)method.Serum HsCRP was detected by latex nephelometry method.All statistical analysis was performed with Windows SPSS 11.5.Results:(1)Compared with the control group,the concentration of 8-isoprostaglandinF2αand HsCRP increased in T2DM group(p<0.01), and it increased more significantly in early nephropathy group(p<0.01). (2)Compared with the control group,the concentration of adiponectin decreased in T2DM group(p<0.01),and it decreased more significantly in early nephropathy group(p<0.01).(3)Serum 8-isoprostaglandinF2α concentration was positively correlated with HsCRP and negatively correlated with adiponectin(p<0.01).Serum HsCRP was negatively correlated with adiponectin.Conclusion:(1)Oxidative stress may be involved in the early nephropathy of type 2 diabetes.(2)Oxidative stress and inflammatory may intereact with each other and lead to type 2 diabetes.But weather there is a causal relationship between them remains unknown.(3)Combined detection of 8-isoprostaglandinF2α,HsCRP and adiponectin may be helpful for the diagnosis of the early nephropathy in type 2 diabetes. Objective:To analyze adiponectin gene SNP+45(T/G)polymorphism of Kunming Han nationality distribution,and to investigate the association of adiponectin gene SNP+45(T/G)polymorphism with type 2 diabetes in Kunming Han nationality. Methods:120 cases of Kunming Han patients with type 2 diabetes and 60 normal healthy controls were randomly collected.The diabetes diagnostic criteria was in accordance with the 1999 WHO diagnostic criteria for diabetes. Genomic DNA extraction was to choose the conventional phenol chloride-ethanol precipitation method.Polymerase chain reaction-restriction fragment length polymorphism method was applied to test the adiponectin SNP+45(T/G)polymorphism.Parts of randomly selected samples were applied by directing sequence to verify the results. Results:120 cases of T2DM patients and 60 healthy controls were for adiponectin genotyping.The allele frequencies were consistent with Hardy-Weinberg balance.(1)Adiponectin gene SNP+45(T/G)polymorphism between type 2 diabetes group and healthy control group had significant differences(p<0.01).The frequency of GG genotype increased significantly in patients with type 2 diabetes.(2)Clinical data analysis of type 2 diabetes group and the healthy control group:age,sex and body mass index were matched,and the leyel of uric acid and total cholesterol had no statistical differences(p>0.05).(3)Fasting blood glucose,insulin, glycosylated hemoglobin,triglycerides and insulin resistance index of T2DM group were significantly higher than the normal control group,and there were statistical differences significantly(p<0.01).The level of high density lipoprotein in type 2 diabetes group was significantly lower than the control group,and there were statistically significant differences (p<0.01).(4)Clinical data analysis of different genotypes in T2DM group:①age,sex,fasting plasma glucose,glycosylated hemoglobin, triglyceride,total cholesterol and low-density lipoprotein had no statistically differences.②fasting insulin:TT group was significantly lower than the GG group,and there were significant differences(p<0.01). There were no significant differences between TG group and GG group. TG group was higher than TT group,and there were statistical differences(p<0.01).(3)high-density lipoprotein:TT group was significantly higher than the GG group,and there were significant differences(p<0.01).TG group was significantly higher than GG group(p<0.01).There were no significant differences between TG group and TT group.④HOMA-IR:GG group was significantly higher than TT group,and there were significant differences(p<0.01).GG group was significantly higher than TG group,and there were significant differences (p<0.01).There were no significant differences between TG group and TT group. Conclusion:Adiponectin gene SNP+45(T/G)polymorphism may be associated with type 2 diabetes in Kunming Han nationality,and GG genotype may be a genetic risk factor of type 2 diabetes in Kunming Han nationality.Objective:To investigate the relationship between adiponectin gene SNP+45(T/G) polymorphism and oxidative stress in type 2 diabetes.Methods:The adiponectin gene SNP+45(T/G)polymorphism analysis of the selected 120 cases of T2DM patients had finished in the second part of the thesis.We detected serum 8-isoprostaglandinF2αconcentrations by enzyme-linked immunosorbent assay method.By detecting serum 8-isoprostaglandianF2αconcentration of different genotypes of adiponectin,We wanted to investigate the relationship between adiponectin gene SNP+45(T/G)polymorphism and oxidative stress in type 2 diabetes.Results:There was no statistic difference of the serum 8-isoprostaglandinF2α concentration in adiponectin SNP45 site of GG,TG and TT genotype in type 2 diabetes.Conclusion:There may be not significant relevance between adiponectin gene SNP +45(T/G)polymorphism and oxidative stress in type 2 diabetes. Adiponectin gene may not be an oxidative stress-related gene.
