| OBJECTIVE: To investigate the interaction between environmental and lifestyle risk factors of type 2 diabetes mellitus in China and adiponectin gene rs10937273,rs1501299,rs182052,rs2241767,rs266729.Methods: The case group was obtained from the Affiliated Hospital of Inner Mongolia Medical University,Zhongmeng Hospital of Inner Mongolia Autonomous Region and Inner Mongolia International Mongolian Hospital.A total of 203 patients were diagnosed.The control group was obtained from 203 cases of the Medical Center of the Affiliated Hospital of Inner Mongolia Medical University.The polymorphism of adiponectin(ADIPOQ)gene rs10937273,rs1501299,rs182052,rs2241767,rs266729 loci was detected by imLDR technique.Generalized multi-factor dimensionality reduction(GMDR)was used to analyze the association between adiponectin gene polymorphism and type 2 diabetes,and to analyze gene-gene,gene and environmental interactions.Results: The ADIPOQ gene rs10937273,rs1501299,rs182052,rs2241767,rs266729 polymorphism allele frequency distribution accorded with Hardy-Weinberg’s law HWE-P values were 0.73,0,6,0.94,0.59,0.47;gene polymorphism sites Correlation with type 2 diabetes resulted in rs1501299,TT vs.GG(χ~2=5.238,P=0.022)and TT vs.TG+GG(χ~2=6.377,P=0.012).The linkage disequilibrium and haplotype analysis of 5 SNP loci of adiponectin gene were obtained: GGAAC was T2 DM susceptible haplotype(χ~2=14.529,P<0.001);interaction analysis showed that: rs10937273,rs1501299,rs182052 There was no interaction between rs2241767 and rs266729;rs1501299 had an interaction with hypertension history and high-fat diet(χ~2=14.529,P<0.001,OR=6.723).Conclusion: rs1501299 is associated with the pathogenesis of type 2 diabetes;G G A A C is a T2 DM susceptible haplotype;rs1501299 gene polymorphism interacts with hypertension history and high-fat diet. |
