Pueraria lobata is the flowers of either pueraria lobata(Willd)Ohwi or P.thomsoii Beeth. In recent years, althought our country has done many researches on pharmaco logy with fruits of Pueraria lobata,The research of chemical component has not been reported systematically. So the purpose of this topic is to study on the isoflavonoids parts of the which are found in the preparative experiments, develop the ground parts of fruits of Pueraria lobata, make a further study on pharmacological effect, and set the basis for the better use and development of this natural resources.This topic adopt method of solvent extraction, then from parts of acetoacetate and n-butanol by silica gel column chromatography and recrystallisation, five constitu ents were isolated. On the basis of spectroscopic analysis methods and chemical meth ods,combinate literature report,identificate four compounds. four compounds were identified as I4',7-dihydroxy-6-methoxyisoflavone,â…¡4',5,7-trihydroxy-6-methoxyisof lavone-7-O-β-D-glucopyranoside,â…¢4',5,7-trihydroxy-6-methoxyisoflavone-7-O-β-D-xylopyranosyl-(1â†'6)-β-D - glucopyranoside,â…£Puerarin.Using the method of ultrasonic- assisted extraction. Concentration and amount of solvent, ultrasonic time, temperature were studied. The best condition was that, the ratio of 70 % ethanol to the Pueraria lobata( w/v)1 :20,extracting 30 min at 80℃,the content of total flavonoids was 5.19%. To inverstigate the technology for purification total flavonoids from pueraria lobata with different types of macroporous resin,the static capacity adsorption, static elution ratio of 5 types of macroporous resin were studied respectively and compared to find the optimum one, and content of total flavo noids were researched and compared to obtain the optimum technological parameters. the AB-8 type of macroporous resin possessed optimum adsorption and elution param eters ,the concentration of Artemisia argyi sample extract was 2.84 mg·mL-1 and its pH was 4.0,after being eluted with 3 ml·min-1of 70% ethol,the yield of total flavornoi ds was 82% and the product content was 64%.Performed the anti-oxidant capability of extraction by three diffierent free radical systems such as DPPH free radical(DPPH·), superoxide anion free radical(O2-), hydro xide free radical(OH·)and kit of total anti-oxidant capability(T-AOC).Result indicated that the extraction had the strong anti -oxidant capability. Studies on activity of protec ting liver and decreasing value of GPT. Statistically, extraction displayed remarkable a ctivity of protecting liver and decreasing value of GPT on rat liver trauma by CCl4.
