Pueraria lobata is widely distributed in our country and the planting pattern includes wild and artificial cultivation, which belongs to the drying root of kudzu. As one of the commonly used chinese medicine, it is proved that it has promoting the living body metabolism, antioxidant, anti-tumor, reducing blood sugar levels, enhancing the body immunity, slowing down aging and some other pharmacological effects. Recently, the active component of pueraria lobata and its pharmacological research and development has been paid more attention. This paper summarizes the chemical components and pharmacological effects of pueraria lobata, and make the qinling mountain pueraria lobata as raw materials to further research the active components of pueraria lobata.In this experiment, the raw materials were refluxed with ethanol to get the extractum. Then, the extractum was extracted with water saturation of ethyl acetate and of n-butyl alcohol. Subsequently, using column chromatography, thin layer chromatography and preparative chromatography to separate the two parts of extraction layer and nine kinds of pure substance were obtained. The structures of compounds were determined by using the one-dimensional 1H-NMR,13C-NMR and the two-dimensional’H-’H COSY, HSQC and HMBC. Finally, six kinds of compounds were determined, they were lupenone, heneicosanoic acid, daidzein, ononin, daidzin, and a new kind of compound. This paper is divided into the following three parts:1. Review. This part introduces the distribution and the main chemical composition of pueraria lobata, such as isoflavone, triterpene, puerarin glycosides, coumarin, starch of pueraria lobata, and its pharmacological effects, such as antioxidant, anti-free radical action, anti-tumor, estrogen effect, protection liver therapy effect, prevention and amendment of cardiovascular disease were summarized. As a kind of traditional Chinese herbal medicine, it is applied in the medical prescription and industrial raw materials largely.2. The extraction and separation of pueraria lobata chemical composition. Make the qinling mountain wild pueraria lobata as raw materials, smashed and refluxed with ethanol to obtain the crude extraction. Then, the extraction was extracted with water saturation of ethyl acetate and of n-butyl alcohol and its chemical composition can be divided into two parts. In the experiments, the open column chromatography, the preparation of thin-layer chromatography, and low-voltage preparative chromatography were used to separate the chemical composition of pueraria lobata. Nine kinds of pure substance were extraced from the ethyl acetate extract of radix puerariae part and n-butanol extract part.3. Identification the chemical structures of compounds from pueraria lobata. Using one-dimensional NMR such as 1H-NMR,13CNMR two-dimensional NMR such as 1H-1H COSY, HSQC and HMBC to determine the structures of compounds. The structures of six compounds were elucidated as lupenone (comp.1), heneicosanoic (comp.2), daidzein (comp. 3), ononin (comp.4), daidzin (comp.5), and a new compound (comp.6).
