Objective: To compare the differences among water-miscible total proteins of velvet, Sika antlerbase and deer bone.We will isolate and purify the special proteins of Sika antlerbase for further study on physical and chemical properties and cell activities of theirs.Method: By the technology of hollow fiber-membrance to isolate and extract the water-miscible total proteins of velvet, Sika antlerbase and deer bone; using SDS-PAGE to make comparasion of the differences among water-miscible total proteins in velvet, Sika antlerbase and deer bone; extracting and purifying the special proteins of Sika antlerbase by means of ultrafiltration, ionic exchange chromatography, gel filtration chromatography, modern biological technology and so on; using cell culture and mass spectrum to study their properties.Results: 1.The yield is 17.1% for water-miscible total proteins in velvet, 8% for water-miscible total proteins in Sika antlerbase, 0.1% for water-miscible total proteins in deer bone by the isolating technology of hollow fiber-membrance.2.There are extremely obvious differences among water-miscible total proteins in velvet, Sika antlerbase and deer bone.Overall 17KD protein is discovered to be a kind of special proteins in Sika antlerbase.3. The 17KD protein was firstly isolated and purified from Sika antlerbase. The 17KD protein isolated and purified from Sika antlerbase attains the electrophoresis and HPLC purity after the examination.4. We have made a research on cell activities of the 17KD protein isolated and purified firstly from Sika antlerbase, which finally indicated that the 17KD protein in Sika antlerbase has obvious multiplicational influences on Vero and L929 cell.Conclusion: 1.The study shows the differences among water-miscible total proteins in velvet, Sika antlerbase and deer bone.Therefore,SDS-PAGE is an effective, fast, stable and simple method that can distinguish velvet powder, Sika antlerbase powder and deer bone powder.2. The 17KD protein was among the special proteins in Sika antlerbase and has obvious multiplicational influences on Vero and L929 cell. The reserch has laid a foundation for further study of the Sika antlerbase's pharmacology activities and the development of new medicines.
