Objective This study intends to ligate left atrial appendage to simulate the Percutaneors left atrial appendage trancatherter occlusion (PLAATO).Then test this method how to effects the body's blood coagulation,fibrinolysis system and atrial natriuretic peptide secretion levels. Select five specific indexes of the Prethrombotic state(PTS): von Wilebrand factor (vWF),P -select element (GMP-140),Plasma tissue plasminogen activator (t-PA),Plasminogen activator inhibitor ( PAI),Plasma protein C antigen (PC),Brain natriuretic peptide (BNP). Discusses the changes of the various indexes in plasma after ligate rabbit left atrial appendage.Methods 50 New Zealand rabbits as experimental animals. Male and female are not strictly restricted; the weight of the each rabbit is about 5±1.5kg. Randomly selected 25 rabbits as experimental group (were performed with open thoracic operation to ligate left atrial appendage under local anesthesia); others as control group (were performed with open thoracic operation not to ligate left atrial appendage under local anesthesia). After a 24-hour operation, experimental group rabbits and control group rabbits are all phlebotomize. To applicate enzyme linked immunosorbent assay(ELISA) to detect the change of 6 indexes : GMP -140,t-pa,PAI,PC,vWF and BNP.Result Compared to the Control group, The level of the GMP - 140,BNP,PC,vWF are all raised,While The level of PAI,t-pa have reduced in postoperative 24h. But no significant difference (P > 0.05).The reasons of the BNP levels rise may be due to the experimental group has larger damage to the cardiac muscle. After a 24-hour operation, the rabbits of the experimental group have done the pathological section of left atrial appendage. The results demonstrate that myocardial cell has not obviously damaged. Conclusion Ligated left atrial appendage were not increase the damage of endothelial cells and platelets activation .Postoperative observation without severe complication such as embolism and hemolysis. The pathological section is also demonstrated that myocardial cell has not obviously damaged. All these indexes illustrate that ligate rabbit's left atrial appendage had no significant effect on coagulation and fibrinolysis system and endocrine activities.
