| Objective:Measure fasting blood glucose, fasting insulin, triglyceride, free cholesterol, total cholesterol, low density lipoprotein, high density lipoprotein and so on to observe the influence of extraction of Chinese goldthread rhizome and balsam pear to experimental 2 diabetes rats'glucose metabolism and lipid metabolism.Method:Feed 10 of 45 Wistar rats at random ordinarily as control group, other 35 rats are used in the model preparation. Make a high fat diet and 1g intralipid for 6 weeks to induce the 35 rats insulin resistance, then inject Streptozotocin at intravenous to induce them Type2 diabet, afterwards divide the successful modeling rats into Type2 diabet model group (physiological saline 50g·kg-1·d-1), the Chinese goldthread rhizome balsam pear extraction treatment group (150mg Chinese goldthread rhizome extraction and 30mg balsam pear extraction·kg-1·d-1), Metformin treatment group (Metformin 100mg·kg-1·d-1) by weight at random. Lg control group (physiological saline 50g·kg-1·d-1) and other groups introduced above for 8 weeks. Make all groups fasting for 12 hours and kill them by femoral artery bleeding. Collect blood to measure related indicators of blood glucose, lipid and insulin. Separate pancreas from the rats and immunohistochemistry islet B cells to observe.Results:The Chinese goldthread rhizome balsam pear extraction group's fasting blood glucose, insulin resistance index, triglyceride, free cholesterol, total cholesterol, low density lipoprotein reduce obviously. the function of isletβcells increases obviously. The pictures of immunohistochemistry islet B cells show that insulin staining positiveβcells of Chinese goldthread rhizome balsam pear extraction group are more than Type2 diabet model group's.Conclusion:Chinese goldthread rhizome balsam pear extraction can improve 2 diabetes blood glucose metabolism and Lipid metabolism and reduce insulin resistance effectively. |
