Objective:To investigate reversal effect of LY294002 (the specific inhibitor of the signaling pathway PI3K/Akt) on Multidrug Resistance Breast Cancer Cell MCF-7/ADR and its mechanism.Methods:the human breast cancer cell lineMCF-7 and the human multidrug-resistant brest cancer line MCF-7/ADR were to be effected by adriamycin(ADM0.1,0.2,0.4,0.8μmol/L) and jointly with LY294002(0.1μmol/L,2.5μmol/L,5μmol/L, 10μmol/L). Drug sensitivity was measured by MTT. Growth cycle and apoptosis of above mentioned group cells were performed by flow cytometry (FCM)tResults:①There was no obvious cell toxicity of LY294002 to MCF-7 and MCF-7/ADR cell, when the concentration of LY294002 was 10μmol/L below.②IC50 of ADM to MCF-7 and MCF-7/ADR cell were 0.14±0.02 and 15.74±0.08 respectively, drug-resistant index of MCF-7/ADR was 112.③LY294002 could increase cell toxic effect of ADM after treated with LY294002(0.1,2.5,5.0,10μmol/L) respectively to MCF-7/ADR for 48 hours, IC50 respectively declined to 13.53±0.10,10.24±0.08,5.53±0.16,2.29±0.12 (P<0.01)④LY294002 significantly increased the rate of apoptosis(P<0.01).The influence of growth cycle was not obvious.Conclusion:LY294002 was able to enhance the depressant effect of the chemotherapy effect to both the two types of the cell, and could partly reverse multidrug-resistant of MCF-7/ADR.
