The extraction, purification and determination of total flavones in Pawpaw were studied. At the same time, the free radical scavenging activity of the total flavones and rutin were also investigated primarily; then 26 trace elements were determineddirectly by ICP-MS.(1) Extraction of total flavones. Ultrasonic-assisted extraction and microwave-assisted extraction were studied systematically.And the best extraction condition of ultrasonic-assisted extraction, was obtained by the orthogonal test, with the yield of 10.54%.(2) Purification of the total flavones. Five kinds of macroporous resin were studied systematically. Total flavones in Pawpaw were absorbed and desorped well by S-8 macroporous resin. Concentration of 1.50mg/mL extraction solution was added into the column with the rate of 1BV/h up to resin being saturated absolutely. Total flavone was eluted by 60%(v/v) ethanol with the rate of 1BV/h. The purity of total flavone was 88.23%.(3) Separation of rutin.Two methods, the polyamide column chromatographic and High-Speed Counter-Current Chromatography (HSCCC) were used to separate rutin in Pawpaw. And the best separation condition was obtained. High-SpeedCounter-CurrentChromatograph was more effective methods, and with the purity of rutin was 97%.(4) Determination of the total flavones and rutin. The total flavones in Pawpaw were determined by spectrophotometry with rutin as standard control. The equation of linear regression was A=0.0096C+0.0094 (R=0.9991), with a good linear relationship in the range of 8~64μg/mL. High performance liquid chromatography (HPLC) was developed to determine trutin, and the optimal conditions of separation and detection were achieved on a Lichrospher C18 column (150mm×4.6mm,5μm) with the mobile phase of CH3CN-H2O-CH3COOH, detected at 360 nm. The flow rate was 0.8 mL/min. The equation of linear regression was Y= 7.7×102+7.0×103×C (r=0.99995), with a good linear relationship in the range of 25~125μg/mL.(5) The antioxidation activities of rutin and flavonoid extract from Pawpaw, the DPPH radical scavenging effect, were investigated.Pawpaw extracts showed good activities, and the antioxidation ability of extracts with different solvents was in the order of methanol extract>ethanol extract>ethyl acetate extract>distilled water extract>chloroform extract. And rutin also had antioxidation activity.(6) Determination of multiplex trace elements.To establish a ICP-MS method for the determination of multiplex trace elements in Pawpaw. 26 trace elements were simultaneously analyzed by ICP-MS. Result was that for all of the analyzed elements, the relative standard deviations (RSD) were less than 2% and the recoveries were in the range of 96.5%~105.6%.
