Objective:To investigate the toxic effects symptom spectrum of Radix Sophorae Tonkinensis and rules of toxic effect, possible target organs toxicity and mechanism. Method:1.Aacute systemic toxicity test:The water extract tonkinensis,70% ethanol extract and total alkaloids. gastric perfusion one-time grant kunming mice, Observe within 14 days of the mice toxic reaction and death。2. Repeated dose toxicity study:The water extract of Radix Sophorae Tonkinensis different doses (Recommended for pharmacopoeia respectively, Clinical dosage of 120,60,30 times) Gastric lavage rats for 90 days and then Withdrawal of 15 days, to observe the long-term toxicity in rats.3. The toxicity study target organs:Key observe the central nervous system of Toxicity, to observe nikethamide and Madopar about the effects of Radix Sophorae Tonkinensis's poisoning, Single and multiple times gives Radix Sophorae Tonkinensis's water extracts and total alkaloid content of mice brain DA, Ach. Result:1. Acute toxicity tests:All three sample of Radix Sophorae Tonkinensis's toxic symptoms spectrum are appear temporary excitement after use the drug.(the alkaloids of Radix Sophorae Tonkinensis About 2-3 minutes appears, Radix Sophorae Tonkinensis's water extracts probably in 10 minutes, Alcohol extract in 10-20 minutes). Then repose, listlessness, less anorexia, close eyes, and then there breathing slow deepen and difficulty to walk, unsteady gait, limb tic, tremor, piloerection, cyanosis etc, until death. Radix Sophorae Tonkinensis's water extract LD50 is 31.66 (原生药) g/kg, Radix Sophorae Tonkinensis's Alcohol extract LD50 is 79.58 (原生药) g/kg, the alkaloids of Radix Sophorae Tonkinensis's LD50 is 52.45 (原生药) g/kg. 2. Long-term toxicity test high-dosage groups on the third day, animal weight decreased significantly,reduce food intake, Article 5 weeks appear different degree of trembling, unsteady gait, and other symptoms.1 months hematology and blood biochemical indicators, viscera coefficient and blank control compare that, TG and Na reduced,CHOL and CK increased, Kidney, adrenals, testicular, epididymal index increased; Compared with the blank control 3 months, no significant changes, hematology index. in the biochemical blood CHOL ALT, increased significantly, viscera liver, lungs, kidneys, adrenals, normalizing, epididymal index increased. There is no abnormal change pathologically. After discontinuation, the rat of Large doses groups weight is increased significantly and liver index reduced, and other indicators with no significant change with the blank control compared.3.The toxicity study target organs:nikethamide can significantly prolongs survival time of animals; Madopar can obviously reduce animal trembling, unsteady gait, convulsions, etc of symptoms, still can increase the survival rate. DA content no significant changes, reduced Ach. Conclusion:1. Radix Sophorae Tonkinensis has the gastrointestinal, nerve, respiratory system toxicity and liver toxicity, toxicity is reversible after withdrawal can restore.2. Radix Sophorae Tonkinensis can inhibit respiratorycenter, can cause drug-induced Parkinson syndrome.3 the toxic substances is it contained alkaloids.4 it's alkaloid may have DA receptor blockade effect.
