| Objective To investigate the effect of tissue inhibitor of matrixmetalloproteinase-2 (TIMP-2) on expression of collagen type I and fibronectin of extracellular matrix in the posterior sclera of the form-deprivation myopia guinea pig.Methods The form-deprivation myopia of guinea pig was induced by translucent goggles. Thirty-six pigmented guinea pigs were randomly assigned to three experimental groups:TIMP-2 group, empty plasmid group and saline group (12 in each group). Right eyes were given treatment, while lift eyes were self-control group without any treatment. Another twelve guinea pigs were selected as control group covering the right eyes all the time. Diopter and eyeball axis was detected at 2th,7th and 14th days after drug injection, after which the guinea pig were sacrificed and the posterior sclera of the eyeball were taken out. The expression of collagen type I and fibronectin mRNA were investigated by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR).Results The mRNA expression level of Col-â… in the posterior sclera of guinea pig was reduced in TIMP-2 group as compared with its self-control group, but the FN mRNA was increased, which showed significant difference (P<0.05). The mRNA expression level of Col-â… began to increase from the 2th day after drug injection. The level was obviously increased at 7th and then decreased at 14th day, which show significant difference as compared with the other three groups(P<0.05). The mRNA expression level of FN began to reduce from the 2th day. The tendency of this went on obviously at 7th and then increased at 14th day. Significant difference was found between them and the other three groups (P<0.05). The mRNA expression level of Col-â… and FN at 7th after drug injection were significant different from that at 2th and 14th days (P<0.01),while, the mRNA expression level of them show no significant different between 2th and 14th days (P>0.05).Conclusion The mRNA expression level of Col-â… in the posterior sclera of guinea pig had dynamic changes as time prolonged after drug injection. It increased tempererily and then began to descend, in the FN mRNA, it had contrary results. TIMP-2 could up-regulate the expression of Col-â… mRNA and down-regulate the expression of FN mRNA. It may slow down the sclera remodeling of FDM guinea pig in the early stage. |
