ObjectiveTo study the isolation, culture, proliferation and collagen production of adipose derived stem cells, and effects of TGF-β1 on cell proliferation and collagen production by this kind of stem cells.MetrhodsThe cells were isolated from SD rat adipose tissue and cultured. The morphologhy of MSCs and their growth characteristics in vitro were observed under inverted phase contrast microscope. The cell passage and proliferation was observed by drawing the cell growth curve. The surface antigen phenotype was analyzed by flow cytometry.Cell culture medium was supplemented with 5μg/L and 10μg/L of TGF-β1. Cell number and collagen I production were measured and compared with unsupplemented control culture and different potency culture.ResultsMSCs could be isolated from adipose. The success rate of isolating MSCs from adipose was up to 58.3%; through the observation under the microscopic, we found that AMSCs were adherent to the walls of the bottles, like the fibroblast morphology and grew like whirlpool. The growth curves of primary AMSCs were"S"type, so were the subculturing AMSCs. The greatest reproductive activity was found in the third and fifth generation of AMSCs. Significant difference was not found in the effects of TGF-β1 on cell proliferation All adipose derived stem cells produced collagen I, There was a significant increase (P<0.05) in collagen I production. Flow cytometry showed that these cells were positive for CD29,CD44 and CD90 expression, but they failed to express hematopoictic cell surface markers, such as CD34 and CD45.ConclusionWe could isolate MSCs from adipose and the success rate is high; the AMSCs have the same biological characters with fibroblast; besides their morphological characters, biological characteristics and cell surface markers are similar with MSCs derived from bone marrow; the capability of proliferation and regeneration is great. Modulation of TGF-β1 levels may modulate collagen production in adipose derived stem cells, which may provide a novel approach to improve tendon healing and prevent adhesion of tendon.
