Purpose: To explore the promoting proliferation and inhibition effect to apoptosis of osteoarthritis chondrocyte of pilose antler polypeptides.Method: 20 Japanese white rabbits were selected into 5 groups at random with 4 in each one. Two of them were made osteoarthritis model by Hulth method and the others performed with sham operation. Chondrocyte separated from double stifle in sham operation animals were taken as control group. Chondrocyte of model group and low dose group, middle dose group as well as high dose group of pilose antler polypeptides were separated from model animals. 10% IMDM were given to the control and model group. Pilose antler polypeptides with three different concentration at 6.25μg/mL, 12.5μg/mL and 25μg/mL were given to the low, middle and high dose groups to culture for a period. The proliferation of chondrocyte was determined by MIT method, the cell cycle as well as apoptosis by flow cytometry.Result: Pilose antler polypeptides have no significant promotion effect to the proliferation and no significant regulatory effect to each phase of cell cycle, but obvious inhibition to early apoptosis of osteoarthritis chondrocyte and protection to osteocyte.Conclusion: The expression of chondrocyte normal morphology is the foundation of stable cartilage matrix, and the abnormal functions of chondrocyte are the beginning and key role of the osteoarthritis changing. In this research, osteoarthritis chondrocyte were obtained from the rabbits osteoarthritis models, intervened with pilose antler polypeptides on different concentrations. The results show pilose antler polypeptides have no significant promotion effect to the proliferation of osteoarthritis chondrocyte. The promotion proliferation effect may be covered up by some factors, which can be further studied. The unsiginificant regulatory effect to each phase of cell cycle and obvious inhibition to early apoptosis of osteoarthritis chondrocyte suggested that pilose antler polypeptides have protection effect to chondrocyte.
