Purpose:To study the therapeutic use of traditional Chinese medicine preparation of Mudan granule on rats with streptozotocin (STZ)-induced diabetes as well as observe the effect on Bcl-2/Bax pancreatic isletsβcells in diabetic rats.Materials and methods:Feed the rats adaptively for a week, and use the STZ diabetic rat model with single injection of chain gland , whose blood glucose is no less than 16.7mmol / L in the model, from which 50 rats were chosen and divided into high dose of wood pellet group, middle dose group wood pellet, wood pellet low-dose group, insulin group, and model group at random. Besides prepare another 10 rats served as normal comparison. Treatment: After the modeling. Normal groups and model groups: lavage with saline 10ml/kg once a day. Lavage with Mudan granule high dose group, middle dose group and low dose group of different doses of once a day: Insulin control group was injected insulin once a day. After 2 weeks, test their weight, and blood glucose. According to the weight, adjust the use of medication, collect the specimen and test their weight and blood glucose after 4 weeks. After the experimental animals in each group were accomplished at the end of the test, that is the rats were anesthetized, and the pancreatic tissue were taken, the following indicators should be measured: the production of light microscope pathological section and dyeing HE; observation of pancreatic tissue morphostructure ; the expression of cell death signals Bcl-2/Bax in the isletβcell with .immunohistochemical detection.Results:1. In the experimental process of general state, inactivity, laziness, polydipsia, polyuria, weight loss, lying curlily, and hair in incompactness and matt appeared. In the model group, while in the Mudan granule low, medium and high dose groups similar situation happened but lower than model group. There was no significant changes in control group and insulin glargine. 2. At the end of the experiment the weight of rats in Mudan treatment group is lower than that in the normal control group, so it is no statistical significance,;P> 0.05,; the blood glucose levels are all higher than those in the control group (P <0.05,)3. Light microscopy: pancreas islets of rats in control group are in the pancreas endocrine whose shape is plump, with a clear outline, and uniform cell distribution in the island; while pancreas islets of rats in model group are significantly atrophy, less rounded profile, and the edge is not correct, Island cytoplasmic reduction of nuclear enrichment and step aside, isletβcells were significantly reduced; glargine group improved significantly compared with model group, island cell distribution rules, pancreaticβwas significantly increased compared with the model; Cells in Mudan middle dose group were round t, and arranged in neat rows than in model group increased pancreaticβ, but less than insulin glargine; low and high dose islet atrophy, outline package is not very smooth, straight edge, nuclear enrichment and step aside.4. The expression of Bcl-2 can be raised and the expression of Bax can be dropped in Mudan granule , which may play a protective role onβcells.Conclusion:Mudan granule can slightly inhibit or delay the Isletβcell apoptosis, which may be up-regulated the expression of Bcl-2 and inhibit the expression of Bax.
