| Objective:This study is to explore the effects of the added Leucine(LEU) and aerobic exercise on the myocardial cell apoptosis in aging mice. Methods:The male ICR mice(13months old) were randomly divided into control group (C),Leucine Group(LEUC), Leucine and swimming training group (LEUE group). CGroup were fed with normal diet, LEUC and LEUE were fed with5%Leucine specialdiet. LEUE group were trained for45minutes per day, five days weekly and a total of8weeks, the mice loaded2.8-3%of body-weight.After8weeks, mice’s hearts wereweighted to get the heart weight index (HWI). take the left ventricle of the heartmuscle, General HE staining, TUNEL detected myocardial cell apoptosis and Elisadetected IBcl-2and Bax protein. Results:1.15age visible in mouse myocardiumpathological changes, characterized by a small amount of myocardial necrosis withfibrous tissue hyperplasia, cardiac myocyte degeneration, interstitial fibrosis,myocardial void degeneration. Severity of myocardial lesions: LEUE groups less thanLEUC group less than the C group. Compared with C group, HWI of LEUC groupand LEUE group were significantly decreased (LEUE groups less than LEUC groupless than the C group), but the differences between two groups is no significant.2.Myocardial cell apoptosis died index (AI): LEUE group less than LEUC group lessthan C group, three groups appear significant differences.3. Expression of Bcl-2protein: LEUE group more than LEUC group more than C group, expression of Baxprotein: LEUE group less than LEUC group less than C group but there was nosignificant difference between the groups. Conclusions: Leucine can reducemyocardial cell apoptosis in aging mice and severity of myocardial lesions. Combinedwith aerobic exercise, Leucine has better effect on alleviating senescencecardiomyocyte apoptosis and myocardial lesions in mice. |
