Ultrafine fibers prepared by electrospinning has the properties of smaller diameter, higher surface and higher porosity, so they can attract vast attention for widely used in various fields especially in the field of biological medicine such as tissue engineering scaffolds, drug delivery carrier and so on. Now with the development of electrospinning technique, electrospun ultrafine fibers as carrier of drug controlled release has had certain development, which is a new thesis in medicine material fields.This paper made PLA with the property of biocompatibility as the drug carrier and mixed with anticoagulant curcumin via electrospinning. In order to study the influence of the main electrospinning parameters on fibers, scanning electron microscopy (SEM) and Photoshop software were used to analyse the surface morphology and diameter of electrospun fibers prepared from different electrospinning parameters; The blood compatibility of electrospun PLA/curcumin composite membranes with different curcumin concentrations was investigated by experiments of platelet adhesion, activated partial thrombin time (APTT) and prothrombin time (PT); Medical Statistical Analysis System (SAS) was applied to identify the main influential factor of APTT and PT experiment and observe if the effect of curcumin had statistical significant according to the results of APTT and PT; The basic performance tests of PLA/curcumin composite membrane were carried out before the study of its drug and degradation property. The addition of curcumin had effect on the microstructure of electrospun composite fibers, which was also tested and analyzed through Fourier Transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD). Wetting property, pore size distribution and mechanical property tests were carried out using our laboratory related equipment for electrospun pure PLA and PLA/curcumin composite membranes with 3% curcumin; UV-spectrophotometer was used to characterize the drug release of PLA/curcumin composite membrane by means of in vitro release,at the same time the degradation property of the electrospun pure PLA and PLA/curcumin composite membranes was also studied by simulating in vivo environment. The results showed that:(1) The main electrospinning parameters all have effect on the microstructure and diameter of electrospun fibers. It shows that: in this experimental research scope, the optimal concentration of PLA is 9 %, the concentrations of curcumin (1 %-5 %) and electrospinnig voltages (12 kV-24 kV) both have significant effect, while electrospinnig C-SD (10 cm-16 cm) and speeds (0.3 ml/h-0.9 ml /h) have less effect;(2) Blood compatibility experiments illustrates that the anticoagulant property of electrospun PLA/curcumin composite membrane is better than that of electrospun pure PLA membrane, and curcumin can significantly control the platelets adherent with 62.78 % inhibition rate of platelet aggregation and increase the APTT and PT, moreover, the anticoagulant effect of composite membrane increases with the increase of curcumin concentration within the range of experimental concentrations; According to the results of SAS, the main influential factor of APTT and PT experiment is curcumin concentration, and the effect of curcumin has statistical significant, meanwhile the statistical significance is more and more significant with the increase of curcumin concentration; (3) Curcumin is well mixed with PLA just by physical mixing instead of the combination of chemical bond, and the addition of curcumin improves the crystalline structure of electrospun membrane;(4) The addition of curcumin improves the wetting property of electrospun composite membrane, meanwhile makes the electrospun PLA/curcumin composite membrane have pores with smaller diameter and narrower pore distribution extending the application in the field of drug controlled release; Compared with the pure PLA membrane, PLA/curcumin composite membrane has higer tensile strength and lower breaking elongation;(5) The accumulative drug release rate of electrospun PLA/curcumin composite membrane increases gradually with prolongation of time, which is different from the lower and relatively steadier accumulative drug release rate of stewing membrane. Electrospun PLA/curcumin composite membrane can maintain better shape with being close to human normal PH without side effect on human; It is found that the drug release and degradation of composite membrance happens simultaneously, and the degradation progress is beneficial to drug release.
