Pyrimidine heterocyclic is in many useful compounds, which used in a lot of Pharmaceuticals, pesticides, supra molecular complexes and natural organic products, etc. Suzuki coupling reaction is widely used in organic synthesis, the response of the classic response in drug synthesis field has produced profound influence. Pyrimidine is an important drug intermediates. In recent years, the study of pyrimidine derivatives was never stoped, especially for the study of Suzuki coupling reaction about this has become hotter and hotter. This paper reviews the ues and synthesis of pyrimidine, and also the suzuki coupling reaction about it. The reports about coupling reaction of pyrimidine with pyridineboronate esters is not lack. This paper selected the chlorinated pyrimidine compounds as the substrate, studied the Suzuki coupling reaction of its and pyridineboronate esters.We synthetized several single chloropyrimidine by used simple materials as the substrate (4-chloro-6-methoxypyrimidin, 4-chloro-2-methylpyrimidine, 4-benzyloxy-6-chloropyrimidine, 6-chloropyrimidin-4-amine, N-benzyl-6- chloro-pyrimidin-4-amine). Used different pyridineboronate esters for relatives nuclear reagent, found that the best conditions about the Suzuki coupling reaction: 2mol%Pd(OAc)2 and 4mol%PPh3 for catalyst, Na2CO3 for alkali, diox and water mixture for solvent, and temperature is 90℃. In this system, the reaction of chlorinated pyrimidine with different pyridineboronate esters can be achieved perfect results, and we also got a series of new coupling products.
