| In this research, the microemulsion of DHA algae oil was prepared by the method of pHase inversion emulsification. The HPLC detection of in the microemulsion and plasma were established. In the screening on the basis of the microemulsion formulation, the factors of formation of DHA microemulsion were invested by the establishment of pseudo-temary diagram. The optimal formula of the high encapsulation efficiency and the large drug loading was finalized. On this basis, the physical and chemical properties and stability of DHA microemulsion were studied.1,The Ultraviolet absorption wavelength, standard curve, recovery and accuracy were studied by HPLC. Results showed that DHA has the maximal absorption in 215 nm. The good linear range of DHA was 14.06-449.92μg/mL.The mean recovery is 101.56%,RSD of the recovery is 2.11%. The results showed that the method was reliable, simple, and reproducible and could be used in the DHA microemulsion determination. The good linear range of the blood plasma was 5.59-357.50μg/mL. The average extraction recoveries of the different concentration were 75.42%, and the average recovery was 99.15%. The RSD of the accuracies with-day was 0.29%.2^ Prescriptions of microemulsion were screened with the HLB and the microemulsion evaluation standard,pseudoternary phase diagrams were used to be observed the formula of microemulsion. The prescriptions of screening were:the surfactant was RH-40:EL-40,Tween80:Span80,RH-40:Span80,EL-40:Span80; the cosurfactant was propanetriol,1,2-propylene glycol,PEG-400 and the absolute ethyl alcohol; The oil phase was IPM and VE. The influence factors inspection result showed that the type and ratio of surfactant and the temperature were the main and the key factors for the formula of microemulsion. the determined of the best formula for the DHA microemulsion were IPM/algae oil/RH-40/Span-80/PEG-400/water,the km=3:1,and the temperature was 75℃.3,In accordance with the optimum formula, the physical and chemical properties of its appearance conductivity,refractive index,viscosity and diameter distribution were measured. The type of DHA microemulsion was O/W,the appearance of DHA microemulsion was light yellow liquid with good flowability; the conductivity,refractive index,viscosity of DHA microemulsion was 116.4μs/cm,1.412 nd,205mpa.s, the morphological character of DHA microemulsion was spheroidal, in uniform distribution, the range of its diameter was 49.5nm.4,The stability of DHA microemulsion was studied through centrifugal stability, and accelerated test. The experiments were conducted by the DHA algal oil, DHA emulsion, adding 0.02% VE DHA oil as antioxidant on the stability of microemulsion. The results showed that the stability of the addition of antioxidants was superior to the microemulsion and algal oil, the general stability of the microemulsion is better than algae oil. |
