Today, people pay more attention to environmental and energy issues. More and more people recognized that hydrogen can alleviate current the two big problems, because the hydrogen has its unique advantage in many new energy. Anaerobic fermentation hydrogen production technology converts biomass energy to hydrogen energy through the microbial metabolism, which belong to green process. It also can achieve efficient use of biomass energy and reduce environmental pollution caused by burning waste biomass, which is consistent with the principle of sustainable development. Therefore, in the long run, anaerobic fermentation hydrogen production technology has wide application prospects. On the base of previous research, this paper considers and optimizes the way of enriching hydrogenogens, and study Physiological Ecology of microbial. Finally, we make a preliminary experiment of hydrogen production by taking cornstalk as raw material.First, we enrich hydrogenogens from the cow dung by heating in the different solution medium, aeration or their combination method. Take the sucrose simulation waste water as the substrate, and use hydrogenogens which are enriched by preconditioning, then study the results of those hydrogen production processes. Optimization of the technology of the preconditioning method by orthogonal experimental design, finally, we get the optimal concentration of bacteria sources though single factor experiment. The experimental results show that the optimum concentration of bacteria source 50 g/L, keep boiling 15 minutes in 0.25% NaOH, then aerate 15 minutes, and the pH of broth is 6, can get better hydrogen-producing effect.Use cow dung which is preconditioned to do experiment of anaerobic fermentation hydrogen production, by taking the sucrose simulation waste water as the substrate. Thought single factor experiment, we get that the optimum concentration of sucrose is 20 g/L. When 4.4g sucrose as reactant, the cumulative yield of hydrogen is 725 mL, the specific hydrogen-producing rate is 164.8 mL/g. When the substrate is cornstalk, the optimum concentration also is 20 g/L, and use 4.4 g cornstalk can produce 493.4mL gas, the specific gas-producing rate is 112.5 mL/g. The specific gas-producing rate changes as concentration of substrate, the highest rate we got are 450.9ml/g and 186.7mL/g.By studying the microorganisms'physiological ecology of fermentation process, we verify that the hydrogenogens are Clostridium genera. Take the sucrose simulation waste water as the substrate, by anglicizing metabolites of fermentation, hydrogen maximum concentration of gas is 60.4%, the main organic liquid product is butyric acid, which is prove that this fermentation process is butyrate-type fermentation. It is also same as the process of Clostridium genera, the utilization of sucrose is 75% in this process.
