Tetracyclines Detection And Pollution Characteristics Analysis On Soil-Manure System Of Pengzhou Vegetable Base

As an indispensable composition, soil plays an important role in human lifes. Livestock and poultry manures have changed from the main organic matter source to the main heavy-metal source in soil.There is growing interest in soil antibiotic pollution, in addition to heavy-metal pollution, chemical fertilizer pollution and pesticide pollution.To study antibiotics behaviors in environment, an efficient detection method must be established. In this paper, a high performance liquid chromatography(HPLC) method was improved based on reported detection methods. Then, this newly-improved method was applied to detect oxytetracycline(OTC), tetracycline(TC) and chlortetracycline(CTC) in Pengzhou vegetable base. Conclusions were summarized as follows:(1)The improved method with higher recovery of OTC, TC, and CTC, were 87.1%, 79% and 85.7%, respectively.(2)The newly-improved detection method was different in SPE activation which used 6mL methanol and 6mL high-purified water; in SPE cleaning which chose 2mL 15% methanol aqueous solution and in SPE elution which was eluted by 3mL methanol. Mobile phase for this method was oxalic acid(0.01mol/L):methyl cyanides:methanol at the ratio of 76:16:8.(3) In Pengzhou vegetable base,TCs (OTC, TC, CTC) were detected in soil, manuer and biogas slurry, at the average detection ratio of 90.7%,81.0%,85.2%, respectively, with their average concentrations 0.452mg/kg,1.766mg/kg,12.265μg/L in response. CTC was at the highest concentration level of 5.325 mg/kg,7.79mg/kg,107.01μg/L in soil, manuer and biogas slurry, respectively. Generally, compared with TCs contents in other domestic areas and abroad, TCs concentrations were lower in soil and manure of Pengzhou vegetable base.(4)From the correlation analysis between soil chemical properties and TCs contents, it could be concluded that, the correlation coefficient between pH and OTC was 0.536**; the correlation coefficient between Cr and TC was -0.401*; the correlation coefficient between OTC and TC was 0.734**; the correlation coefficient between OTC and CTC was 0.682**; the correlation coefficient between CTC and TC was 0.830**.
