Sucralose is a non-sweetener that was synthesized by chloride of sucrose,with the porperty of high sweetness,low heat, orthodox taste,safety and stabilization.have been applied to medicine,health product,and food.At present the market demand of the high-purity sucralose(purity reached 98%) increased in domestic and abroad,and treng of short supply.By comparing the determination method of sucralose,using different methods to purify sucralose(purity reached 90%) and explore process conditions,last part stability of sucralose was discussed in this thesis.Comparison of HPLC and chemical titration.The RSD is 0.28%(n=5)and recovery rate is 98.8%by HPLC,The RSD is 1.48%(n=5) and recovery rate is 90.3%.The measured value of chemical titration above the HPLC's.The purification of sucralose raw material was studied by column chromatography with silica-gel as adsorbents,and the effects of fixation phase,desorption agent and ratio, loading amount and velocity on the purification were also investigated.Through adsorption and desorption experiments,the best experimental condition are obtained as follows:200-300m silica gel was selected as fixed phase,the desorption agent is ehanol/ehyl acetate(1/4,v/v),the loading amount not more then 0.12g sucralose raw material/g silica gel,and the speed of flow is about 1.Oml/min to 1.5ml/min.Gathered up eluent in different section,then vacuum drying.98%sucralose was gained in the process.Extracte solution of sucralose raw material with ehyl acetate as extractant. the best experimental condition are obtained as follows:for 20g sucralose sample,the amount of extractant is 50ml,the extraction time is 20min,the extraction temperature is 30℃,the holding time is 3h,then crystallize of sucralose with ultrasound,98% sucralose was gained in the process.As the influences of ultrasound on crystallization of sucralose. The best crystallization condition are obtained as follows:crystallization temperature is 40℃,ultrasonic frequency is 59kHz,ultrasonic power is 100w.The thermal stability of sucralose crystalline was analyzed by thermo gravimetry/differential thermal analysis and the melting point of sucralose is about 125℃.The stability of sucralose solution was studied,experimental results show that:the sucralose solution is very stability.
