In this thesis,evening primrose oil and fatty acids was the raw material,and urea adduction and silver of the column chromatography was adopted fatty acids.Through observing and analying the differeces between the shape and aperture size of Urea Inclusion Compound formed under diffent design process with microscope,and the differences between the weight of fatty acids under defferent design process with scale,and the composition of fatty acids before and after separation by gas chromatography (GC), research results were as follow.1.The results indicated that the ratio of fatty acids and urea, inclusion of temperature, time of inclusion have great effect on Urea Inclusion Compound. The results showed that the shape of Urea Inclusion Compound was the most complete and the size was the biggest under the conditions of the ratio of fatty acids and urea was 1:3, temperature of inclusion was -15℃, time of inclusion was 24h or 36h. The three factors and its interactions significantly affect the purity of GLA. The results also showed that the ratio of fatty acids and urea was 1:3, temperature of inclusion was -15℃, time of inclusion was 24h were the best process . Under the best condition, the purity of GLA reached up to 95.5759%.And the same result as the inclusion compound. It was feasible to observe the shape and the amount of the urea inclusion compound to reflect and guide the urea inclusion technology.2. Through observing the differeces between the shape and aperture size of Urea Inclusion Compound and inclusion rate of fatty acids,the result was showed as follow. When the proportion of fatty acids and urea was higher, the inclusion rate of Octanoic acid, Lauric acid and Nutmeg acid was increasing, while Hexadecanoic acid was decreasing; With the increase of the ratio of urea and alcohol, the inclusion rate of Octanoic acid and Lauric acid declined, Hexadecanoic acid reducecd ,while Nutmeg acid increased firstly and then decreased; with the increase of alcohol concentration the inclusion rate of four kinds of fatty acids was decrease. Taking the inclusion rate of four kinds of saturated fatty acids into account, it was feasible to carry out urea inclusion test by low concentration of alcohol ,instead of high concentration of alcohol.It could provide a reference for the Urea inclusion process.3.The result of Silver column chromatography was showed.When the unsaturated degree of fatty acids—the number of double bond was increased, the agents polarity of eluent was increased and the concentration of eluent was increased. The saturated fatty acids and monounsaturated fatty acids were first eluted,and then polyunsaturated fatty acids. The elution effect of Petroleum ether– acetone was better than petroleum ether– ether.The purity of GLA from evening primrose oil was reached to 90.60% by silver turn silica gel (100 or 200 mesh) column chromatography with petroleum ether– gradient.
