Organophosphorus compounds have found a wide rang of applications in the areas of industry, agriculture and medicine due to their chemical, biological and physical properties as well as their utility synthetic intermediates. As a kind of natural amino acid analogues,α-amino phosphonate constitute an important class of compounds with diverse biological activity and structure type, and have attracted the attention of chemical and biological researchers about their biological activity and molecular design. In recent years,α-aminophosphonate has been widely used enzyme inhibitors and antibody in enzyme-catalyzed reaction, In addition, they have excellent application to their antiviral, PGR, antimicrobial and anticancer activity. Sinceα-amino phosphonatesynthesis by Fields in 1952, various methodologies have been published. Of these methods, the one-pot reaction, catalyzed by lewis acids base, is the most conventient.In this paper, we reported a microwave-assisted solvent-free and catalyst-free method for the synthesis a new series of bis(α-aminophosphonates), we also can do with more excellent activity compounds introducing different functional groups. On the other side, for the first time, titanocene dichloride is reported as a new and efficient catalyst for the synthesis ofα-aminophosphonates under solvent-free conditions.This thesis includes three chapters:Part ?: Recent advance in biological activity and synthesis ofα-amino phosphonate. This part is divided into four aspects. Firstly, reviewed the development of green synthetic methods. Secondly, the brief application ofα-amino phosphonates in medidine, chemical industry and so on, and reviewed synthesis method at home and abroad in recent years onα-amino phosphonates, this part includes imine addition, mannich-type addition reaction, microwave irradiation reaction, catalyst synthesis etc. Thirdly, through literature review, outlined the research of doubleα-amino phosphonates. Finally, briefed the research ofα-amino phosphonates in the period of graduate.Part II: 1. Green synthesis and fungicidal activity of novel 1,4-phenyl bisaminphosphonates. Efficient solvent-free and catalyst-free method for the synthesis of nine kind 1,4-phenyl bisaminophosphonates are developed by a microwaveassisted three-component Kabachnik-fields reaction. 2. 4,4′- benzidine bisaminphosphonates using microwave irradiation under solvent-free, catalyst-free conditions. Determine their structure.PartⅢ: Titanocene dichloride was used as an extremely efficient catalyst for a three-component one-pot reaction to formα-aminophosphonates under solvent-free conditions at ambient temperature. The major advantages of the present method are good to high yields, short reaction, environmentally friendly.
