It is important to determinate the trace toxic heavy metals in natural waters, soil and air. They are harmful to the health of people.Heavy metal ions were determined by using ion exchange resin and urease. The resin was used for adsorption and concentration of heavy metal ions in water, which would inhibit the urease activity to determine the concentration of metal ions. A theoretical basis was provided for the enrichment, detection integrated calorimetric biosensors. Resin was studied by different heavy metal ions adsorption properties. The standard curve was studied of the metal ion concentration and ammonium ion concentration by using Spectrophotometric method. Relationships such as temperature, resin quality, buffer, urea, ion species, and ion concentration conditions on urease activity inhibition were studied. The results showed: 50ml heavy metal ions concentration of not more than 0.7mg/mL could be completely adsorbed by 0.5g resin; urease was inhibited in certain the concentration range of copper and lead ions.Urease was inhibited by copper ions and urease was inhibited by mixed metal ions which were contrasted. Copper ions had great effect on the inhibitory of urease; Mixed metal ions had poor effect on the inhibitory of urease. Because mixed ion mutual influence their urease inhibitory effect. Urease is highly sensitive to copper ion. The investigation of copper ions on urease inhibition can be measured. The instrument reference to copper ions on the inhibition of urease is a standard to determine the other ions concentration range.The heat of the urease hydrolysis was tested by instrument. It showed:the resin adsorption of copper ions would make the reaction of the urease hydrolysis release less heat. Copper ions were adsorpted by the resin which inhibited the activity of urease. This inhibition can be determined by the values shown obtained from the instrument designed by us. The conclusion of the determination had the same result with the chemical methods.
