4-(4-phenylbutoxy) benzoic acid is the intermediate of Pranlukast for asthma treatment. In the last few years, the number of patients with asthma increased year by year as the environmental deterioration. The prevention and control of asthma is imminent. In this paper, the process using succinic anhydride as the starting material, going through FriedelCraft reaction to get 3-oxo-phenylbutanoil acid, and then Wollf-Kishner reaction to get 4-phenylbutyric acid, and in NaBH4-I2 system to obtain 4-benzene-1-butanol. and by bromination reaction, alcoholysis with methyl parahydroxybenzoats, saponification. At last, the target product 4-(4-phenylbutoxy) benzoic acid was obtained. Synthesis of 4-benzene-l-butanol was emphasized in the paper with an orthogonal test optimum reaction conditions. A method of high performance liquid chromatography (HPLC) with external standard was established to the quantitative analysis of 4-phenylbutyric acid. Optimum conditions were determined which was an important material in the synthesis of 4-(4-phenylbutoxy) benzoic acid.In part two of the paper, the reasch of resolution on 4-chloro-a-hydroxyphenylacetic acid obtained by using chlorobenzene formaldehyde and chloroform as starting materials, TEBA as phase transfer catalyst was studied. The influences of extraction method and the acceleration drops of NaOH solution on the reaction to of response were investigated. The optical activity resolving agent N-Benzyl-a-methylbenzylamine was synthesized by benzene procedures and benzaldehyde. For the resolution of 4-chloro-a-hydroxyphenylacetic acid, the crystallization enantiomeric separation method was adopted in this paper. The influences of different solvents and different Moore ratios to resolving agent and 4-chloro-a-hydroxyphenylacetic acid on the effect of the resolution were inspected.
