The Applied Research Of Screening, Identification, Optimization And Fermentation Conditions Of Fungi To Produce Neutral Cellulase In Textile Industry

We isolated 8 fungi from two kinds of soil sample through the screening by congo red. In this study,8 strains of dominant fungi were selected which have the abilities to degrade cellu-lose. Their CMC activity were 3.49 U/mL; 8.08U/mL; 4.55U/mL; 3.73U/mL; 40.16U/mL; 6.54 U/mL; 3.16U/mL; 4.64U/mL. The number 5 strain has the highest CMC activity of 40.16 U/mL.18SrDNA sequences were obtained by PCR amplification. The number 5 strain was Graphium fructicola after being compared in the database.There was a higher CMC activity in the pH range of 5-7 and a better pH stability of the number 2,3,5 strain. It could keep the CMC activity of 50 percent after 4 hours of incubation. They had the higher CMC activity in the temperature range of 40-60℃. The number 5 strain could keep the CMC activity of 50 percent after 2 hours of incubation in 60℃.The number 5 strain had a higherβ-glucosidase enzyme activity and FPA activity respectively. They were 100.890U/mL and 6.993U/mL.The level of cellulase production was improved by the method of single factor and orth-ogonality. Under the condition:medium composition of 1% bran,1% microcrystalline cellu-lose,2% fiber material 1% peptone,0.7% corn steep liquor,3g/L K2HP04, lg/L MgSO4.7 H2O, the initial pH 8,5.2% inoculation,40℃,200r/min for 96 hours, the cellulase activity a-chieved 608.5 U/mL. In addition, the cellulose produced by the eight fugi have great adva-ntages in the effect of denim three-dimensional, flower effects, flowers dot size and flexibility. The cellulase produced by number 5 strain has a great flower effect. The cellulase produced by number 3,4,8 strain had a strong denim three-dimensional sense. The softness of the cellul-ase produced by number 2,7 strain was better and had delicate flower spots.
