| Curcuminoids are natural pigments in the rhizome of Rhizoma Curcumae Longae,such as Turmeric, Turmeric Root-tuber, Zedoary. Conducting the research on the extraction,refining and analysis methods of Curcuminoids has prodigiouss practical significance.The task has mainly researched the methods of extraction,refining, analysis of Curcuminoids in Turmeric.The best extraction and refining methods of Curcuminoids were determined and the analysis methods of Curcuminoids were set up through experiment research.It could be educed that the acetone-extracting method was the best Curcuminoids extraction method through the method of one-factor experiments,with high extraction ratio,good extraction effect and economic efficiency,favourable security.The method of L9(34)orthogonal experiment has been used to research the best extraction conditions of acetone-extracting method. The effects to extraction ratio of 4 factors—the particle size of Turmeric powder,extraction temperature,ratio of material to extraction solvent and stirring rotating speed have been comprehensively investigated. Eventually,the best extraction conditions of acetone-extracting method could be detemined through analysis to the results of orthogonal experiments and confirmatory experiment—the particle size of Turmeric powder 40,extraction temperature 30℃,ratio of material to extraction solvent 1:10,stirring rotating speed 400r/min.The extraction ratio attained to 97.54% in the best extraction conditions. The best Curcuminoids refining method and the best refining conditions were detemined by the method of one-factor experiments.Three Curcuminoids refining methods—acid-alkali method, organic solvent extracting method and chromatography has been investigated. The experiment results indicated that the best Curcuminoids refining method was activated Clay Column Chromatography.And its best experiment conditions were: the adsorption temperature 15℃,the flow rate of extraction liquid in adsorption phase 2.5mL/min,the elution temperature 25℃,the elution flow rate 2.5mL/min, the elution solvent pH 12.0.On these experiment conditions,the ultimate extraction ratio attained to 85.61%,and the purity of refined product attained to 93.78%.The best Curcuminoids analysis method has been setten up. The Maximal Absorbance Wavelength of Curcuminoids was scaned and detemined by ultraviolet-uisible spectrophotometer at first.Afterwords,two frequently-used Curcuminoids analysismethods—ultraviolet-uisible spectrophotometry and high performance liquid chromatography were investigated. External standard method was used to analyse the content of Curcuminoids and the standard curves were drew out. The experiment results indicated: the Maximal Absorbance Wavelength of Curcuminoids was 420nm, the two analysis methods of Curcuminoids respectively had their relative merits.The former is easy and simple to operate and its analysis way is fast and convient.It's applicable to rapid and rough determination of Curcuminoids content of sample.The latter is relatively complicated to operate,its analysis speed is relatively slow.But its analysis result is accurate and reliable,and it can separate the three different curcumines of Curcuminoids.So it's applicable to strict and quantitave analysis of curcumines content of sample.Through comprehensive comparison,It was determined that high performance liquid chromatography was superior to ultraviolet-uisible spectrophotometry. |
