Isodon excia (Maxin.) Hara, also named Gui Ye Cao,Gou Ri Cao,Ye Su Zi, belongs to Labiatae family and is widely distributed in northeast China, especially in Jilin, Liaoning and Henan Province. In the past few years, the study on chemical constituents and biology activity of Isodon excia was still at an early stage. So far 34 diterpenoids were separated from Isodon excia. Besides the diterpenoids constituents, it contains volatile oil and ursolic acid, daucosterol, liquiritin etc, and some trace elements. People studied on 7 areas of the biology activity of Isodon excia. They are effect of againsting tumour, antibacterial, antiviral and immunoregulation, antioxidation, and on the cardiovascular system.Based on previous studies, same plant in different regions may have different secondary metabolites. So this paper systematically studies on Isodon excia of the Changbai Mountain. The dried leaves of Isodon excia were extracted with solvent by using ultrasonic. Fourteen compounds were isolated by liquid-liquid extraction, MCI column chromatography, normal and reverse column chromatography and semipreparative HPLC. The structures of 5 compounds were established by MS, 1D and 2D NMR analysis. They are euscaphic acid, Pseurata A,1-Hydroxypinoresinol, caffeic acid methyl ester, Glaucocalyxin A. Among them 1-Hydroxypinoresinol had not been determined on the stereostructure. These compounds are isolated from Isodon excia for the first time. The rest compounds will be identified for further analysis.In this paper, all the fractions and compounds were investigated by radical scavenging activity test and tyrosinase inhibitory activity test. The results of activity tests showed that the most of fractions exhibited strong free radical scavenging activity. The inhibition ratios of fractions were over 90%,14 fractions was reached 100%. Only some of the fractions exhibited good tyrosinase inhibitory activity, IE1f2c5 and IE1e2b were reached 89.7%. IE2, IE3, IElf10, IElf2c5 and IEle2b have two kinds of strong activity. The inhibition ratios of fractions were over 80%.14 compounds were isolated by activity test as a guide. Compound 9 and compound 12 have strong free radical scavenging activity, the inhibition rates were 95.4%,90.1%, the IC50 values were 0.01,29.87. Compound 8 and compound 10 exhibited good tyrosinase inhibitory activity, the inhibition rates were 51.5%,55.0%. Antioxidant activity of Isodon excia is still at an early stage, meanwhile the tyrosinase inhibitory activity has not been reported. Our work is to develop new antioxidant and tyrosinase inhibitor drugs to provide the basis of experimental data.
