Aphid is an important pest in agricultural production. Aphids damage the plants through several ways:attack the plants directly; the honeydew secreted by the aphid can breed disease; the major medium between the plants and the plant disease. The concept of pest resist has been changed, from "kill" in the past to "control" now. The aim of control is to control pest efficient but beneficial to environment and ecological balance. So we should take full advantage of the tritrophic system interactions among host crops, pest and predator to control the pest. When attacked by predators or parasitoids, aphids produce (E)-β-farnesene which cause other aphids in the vicinity to stop feeding and to move away. We transferred the (E)-β-farnesene synthase into rice through transgenic technique. The rice can release endogenous (E)-β-farnesene into environment to alarm the aphids. This strategy is good for environment protection, food safety and predator protection.Our experiment linked the (E)-β-farnesene synthase, which cloned from MenthaⅩpiperita, onto the vector pCAMBIA1300s. After transferred the Eβf synthase gene into Zhonghua 11 by Agrobacterium-mediated transformation, we identified the resistance of the transgenic plants. The main results are as follows:1. Eβf synthase gene was carried by pArt7-EBFS originally, and finally linked to pCAMBIA1300s by PCR.2. Transfer the expression vector, which contain Eβf synthase gene, into japonica Zhonghua11 by Agrobacterium-mediated transformation. After selection, differentiation and root, we got 65 transgenic young plants.3. Through PCR, Southern blot, germination experiment and phenotype examine, we got 4 homozygous transgenic families with single copy.4. Detect the repellent of the 4 transgenic families to aphids. The result suggests that the Eβf produced by transgenic rice can disperse the aphids.5. Detect the repellent of the 4 transgenic families to rice brown planthoppers through olfactometer osphresiometer of Y type. There are no significant differences between transgenic and non-transgenic rice on rice brown planthoppers.6. Detect the repellent of the 4 transgenic families to rice stem borers through olfactometer osphresiometer of Y type. There are no significant differences between transgenic and non-transgenic rice on rice stem borers.
