Cymbidium hybridum, also known as Cymbidium hookerianum, is a kind of high-value ornamental cattleya. Cymbidium, with its large flowers, straight branches and spindly leaves, is deeply welcomed by Cymbidium enthusiasts and is also regarded as the upmarket potted flower for Spring Festival consumption. However, because Cymbidium hybridum mainly belongs to hybrid variety, the reproduction of its seeds is unable to maintain its variety characteristics. Morever, its fructification rate is quite low and branching capability is also very weak, all of which results in extremely low reproduction coefficient and tardy reproduction speed. Thus, Cymbidium can hardly satisfy the need of industrial production by far. Due to the inefficiency of its industrial production and the shortage of many precious varieties, the price of Cymbidium hybridum has been climbed unceasingly in recent years. Therefore, the establishment and improvement of Cymbidium hybridum's high frequency regeneration and mass production technology is the key to solving this problem.The new-born lateral buds from Cymbidium hybridum's pseudo bulbs were taken as explants to induce protocorm and cultivate aseptic seedlings. This experiment makes a research on the main factors that effect the adventitious buds'regeneration outside Cymbidium hybridum's lateral buds and establishs the system of Cymbidium Hybridum's high-frequency regeneration, providing technical support and theory basis for Cymbidium hybridum's mass production.The result shows that the most effective procedure of disinfection when taking new-born lateral buds from Cymbidium's pseudo bulbs as explants is as follows:first, clean the gemma of the Cymbidium by tap water; second, dip it in 70% alcohol about 30s; third, dip it in 0.1% HgCl2 for 5mins; then soak it in 500mg/L PVP about 6mins; at last, inoculate it. It is indicated that the contamination rate comes to 18.33%, browning rate 33.33% and survival rate 36.66%.The optimum culture medium compound of inducing protocorms is MS+BA 2.0mg/L+NAA 1.0 mg/L+agar8.0g/L+sugar30g/L+active carbon 2.0g/L, pH5.4. The average induction rate comes to 63.3%.The optimum culture medium compound of protocorm's multiplication isMS+BA1.0mg/L+ NAA 0.5mg/L+IBA 1.0mg/L+agar6g/L+sugar30g/L+potato extract 150g/L+active carbon 2g/L, pH:5.4.The best culture medium for promoting Cymbidium hybridum's seedlings and rooting is 1/2MS+NAA0.1 mg/L+GA31.0mg/L+sugar20g/L+potato extract 100g/L+agar8g/L,pH5.4.The best culture medium for Cymbidium hybridum's seedling by one time is 1/2MS+ BA1.0mg/L+NAAO.1mg/L+potato extract 100g/L+agar 8g/L+sugar 30g/L+active carbon 2.0g/L, pH:5.4.The seedling refining before transplanting can notably increase survival rate of transplant. The optimum duration of seedling refining is 8 days in the room (closed bottle 5 days+opened bottle 3 days). The experiment shows that the full closure can increase survival rate of transplant effectively. Planting material with the merits of keeping wet, warm and ventilative can be regarded as superior soilless media for Cymbidium, which can improve the survival rate to 91.5%.
