Studies On The Comparison Between Different Propagation Ways And Seedling Quality Of Cymbidium Hybridum In Vitro

At present, tissue culture technique is an effective approach to industrialized production of Cymbidium hybridum.Test-tube plantlets of Cymbidium hybridum propagated by tissue culture technique are the main sources of cultivated production of seedlings. In this study, this experiment established the tissue culture system of Cymbidium hybridum through two different propagation ways.One is the protocorm like bodies (PLB) way, the other is cltustered shoots way. Studied the suitable medium for every culture stage, and comparied two different propagation ways various aspects in detail, and provided scientific basis for the actual production.The results showed that, lateral buds are the best explants; two steps (By 10%NaClO sterilized 10min, and then by 5% NaClO sterilized 5min) is the appropriate sterilization way; the optimal induced medium is MS+6-BA2.0mg/L+IBA0.2mg/L.The lateral buds would be induced two times.and it raised the utilization rate of explants. The optimal medium for PLB proliferation and trandformation is MS+KT1.Omg/L+NAA0.1mg/L; the multiplication ratio and trandformation rate are higer. The optimal medium for test-tube plantlet proliferation is MS+6-BA3.0mg/L+2,4-D1.0mg/L+AC1500mg/L.It could keep a higer multiplication ratio and solve the browning problem during test-tube plantlet proliferation.The optimal medium for rooting is 1/2MS+NAA0.05-0.5mg/L+mashed banana 50g/L-100g/L.Compaired the two different propagation ways, the results showed that,the clustered shoots way has higer inducing rate,shorter growth time,higer quality of test-tube plantlets.The quality of test-tube plantlets from clustered is better than test-tube plantlets from PLB way after transplanted.Hence,the clustered shoots way will be the propagation way for industrialized production in future.
