This survey was undertaken based on the investigation on cultivar resources of sweet Osmanthus in Hubei and the south of Jiangsu, Summarizes the situation of the cultivar resources of sweet Osmanthus in the two provinces. Took advantage of ISSR molecular marker, researched the genetic diversity of 40 cultivars of sweet osmanthus, Improved DNA extraction method, established a simple practical ISSR - PCR reaction system, main results are as follow:1.37 cultivars are identified in Hubei province, which belong to four cultivars groups including 3 of Asiaticus Group,18 of Albus Group,8 of Luteus Group and 4 of Aurantiacus Group,each group is formulated a cultivar classification system. Meanwhile,4 confused cultivars are discussed.33 cultivars are described by predecessors. Incorporated Osmanthus fragrans'Zigeng Ziyin' into Osmanthus fragrans'Zi Yingui'. Judging from this survey, there are rich resources of cultivars of sweet osmanthus in Hubei province. Most of them are indigenous tree. Osmanthus fragrans'Jiangnan Liren', Osmanthus fragrans'Yinxing' Osmanthus fragrans 'Boye Yingui', Osmanthus fragrans'Liuye Gui'and Osmanthus fragrans 'Ziyun' which belong to Albus Group; Osmanthus fragrans 'Qiugui'and Osmanthus fragrans 'Fujin' which belong to Luteus Group are all have ancient cultivars. Osmanthus industry has already covered the four group. Osmanthus fragrans 'Jiangnan Liren', Osmanthus fragrans 'Boye Yingui', Osmanthus fragrans 'Yinxing' and Osmanthus fragrans'Xiaohua Jingui'are main cultivar for flower-gathering.2.63cultivars are identified in the south of Jiangsu, which belong to four cultivars groups including 9 of Asiaticus Group,24 of Albus Group,15 of Luteus Group and 13 of Aurantiacus Group,each group is formulated a cultivar classification system. Meanwhile,2 confused cultivars are discussed.61 cultivars are described by predecessors. Incorporated Osmanthus fragrans 'Zhouye sijigui' into Osmanthus fragrans 'Siji gui'.Judging from this survey, there are rich resources of cultivars of sweet osmanthus in the south of Jiangsu. Part of them are indigenous cultivars,but most of them come from Hubei,Zhejiang,Sichuan and Chongqing.The Albus Group is not as much as the same group in Hubei. Osmanthus fragrans'Suzhou Zaohuang', Osmanthus fragrans 'Boye Jingui' are all have ancient trees. Osmanthus industry has already covered the four groups. Most cultivars are widely used in garden.3.There were 11 primers selected from 35 primers, which were applied in genetic diversity and systematic relationships.The number of amplified fragments was 128 while the number of polymorphic fragments was 99.The percentage of polymorphic fragments was 77.34%.Primer UBC825 has the highest percentage of polymorphic fragments,it's 100%, Primer ISSR27 has the lowest percentage of polymorphic fragments,it's 50%.The Primer ISSR12 can indentify the Specific loci of Osmanthus fragrans'Jiulong Gui'.Genetic diversity about the tested materials is rich.4.Using Ntsys-pc version 2.10 get an UPGMA dendrogram. All the materials were divided into 4 groups with the threshold of 0.673,and the cultivars from the same Origin in the same group. The result is almost the same as Classical taxonomy.
