Bambusa multiplex'Fernleaf' is a well known ornamental bamboo. The optimal callus culture medium were screened, and the morphological characters of the callus were observed by SEM.The results were:①The best basic condition for initial culture were:MS medium with 30g·L-1, buds from germfree plantlet as explanta, and Vitamin C 100mg·L-1 as browning-preventative reagent.②The suitable formula for callus induction were:MS+2,4-D 4mg·L-1+6-BA lmg·L-1.After callus induction could get three different types of callus:Type I:This type of callus was light yellow or white, had a smooth and compact surface with a hard texture. They had a low hydrated rate.Type II:This type of callus was white or gray, had a loose and granulated surface with a soft texture. They had a high hydrated rate than type I.Type III:This type of callus was transparent or semitransparent, had a strong stickness and easy to separate. They had a high hydrated rate.Type I was the best callus for proliferation and differentiation.③The suitable formula for callus proliferation were:MS+2,4-D 3mg·L-1+NAA 0.5mg·L-1+6-BA 0.1mg·L-1.④The observation of the different types of callus and isolated culture cell's suface by the SEM were shown:Embryonic callus had a smooth surface and crumb texture with compact combination. Between the crumb was big interspace and combine loosely with regular shape cells. The exposed cells and embryonic cells with paste were observed in the sueface without blanket.Non-embryonic callus had a soft texture and blanket with pieces or snowflake like substances. The shape cells was inregularly with a big volume.Browned callus's surface was very rough and the cells's shape was inregular extremely with much more blanket.Isolated culture cell's suface had particle and fibril.
