Breed is one of the most important factor to restrict our country's layer cultivation.laying performance and egg quality traits not only the most important economic characteristics of chicken, but also has large effects on the economic benefits in egg production. Because of they are low heritability,it's difficult to improve by conventional breeding.The applications of MAS supplied a new method to solve this problem.Xinhua chicken E strain is test materials,PRL gene, VIP gene and FSHβare candidate genes of egg performance.We use PCR-SSCP,sequencing and PCR-RFLP technologies to detect gene polymorphism,filter SNPs and genetyping.At last,we studied different genotype effects with egg performance record through correlation Analysis.The research primarily results as follows:1. We found 5 SNPs in VIP gene,4 SNPs in FSHβgene and validated a 24bp insertion/deletion in PRL 5'regulatory region respectively. Furthermore,we authenticated A-3458G locus of VIP gene, C-358T, T+259G locus of FSHβgene.2. We analysised genetic equilibrium with A-3458G locus of VIP gene,24 bp insertion/deletion locus of PRL gene,C-358T, T+259G locus of FSHβgene.The results of x2 test showed that A-3458G locus of VIP gene fitted Hardy-Weinberg equilibrium,the other three locuses deviated Hardy-Weinberg equilibrium3. A-3458G locus of VIP gene was significnatly(P<0.05) associated with Egg index trait.AA genotype individual outstriped GG genotype individual.we suggested that select individual which include A alleles in breeding.4. T+259G locus of FSHβgene intron 2 was significnatly(P<0.05) associated with egg performances such as age at the first egg,weight of album,percent of album and haugh unit.TT genotype individual outstriped GG genotype individual.We concluded that FSHβgene can be used to carry out MAS breeding as candidate gene. We suggested that select individual which is TT genotype in breeding to improve laying performance and egg quality traits.
