Wheat stripe rust caused by Puccinia striiformis f.sp tritic is one of the most terrible diseases in wheat production. In ordinary year, it can cause reduction by 20-50%, but in extremely prevalent year, it can cause more loss and even no production. For long, the control of wheat stripe rust has depended on the chemical control and the cultivation and promotion of disease-resistant varieties. The loss of resistance to wheat stripe rust of the existing wheat varieties in production practice has been caused by the change of the composition and the frequency of stripe rust fungus. The amount of conidiation production has an effect on the survival capacity, development and popularity of stripe rust fungus to a certain extent. And the conidiation production of stripe rust fungus is influenced by the control of its conidiation-related gene. Therefore, to discover the reasons of different spore production from different races through analyzing the conidiation-related gene PsCon1 of spore production, so as to predict the trend of different micro species is of great importance to the breeding of wheat stripe rust-resistant varieties and the control of stripe rust fungus. As far as observed, there hasn't been any reports about analyzing the conidiation-related gene PsCon1 of different physiological races have almost not been seen till now. As a result, it is significant to clone the serial difference of different conidiation-Related Gene PsCon1 and use the molecular cloning method to predict its trend.It is found that in the reproductive process of wheat stripe rust fungus different physiological races has different conidiation production. PsCon1 was cloned from eight physiological races by molecules cloning method and analyzed the difference in nucleotide sequences and amino acid sequence level so as to reveal the reasons of different conidiation production. The specific research results are as follows:1. Conidiation-related gene PsCon1 was cloned from CY33, Su11-7, CY32, Hybrid 46-8, Sull-3, Su11-4, Su11-5, Su11-13, a part of which registered in the cloning sequence GenBank begins at 83bp and ends at 613, including three exons(79bp-132bp, 213bp-359bp and 432bp-494bp). and two introns (133bp-212bp and 360bp-431bp).2. There was no apparent change whether in the nucleotide sequences or in the amino acid sequences of 8 physiological races. And only some individual site happened mutations. In 45bp, CY 33 and CY 32 have different bases from the other six: the base of CY 33 and CY 32 is C, while the base of the other strains water 11-7, Hybrid 46-8, Sull11-3, 11-4 11-5, and 11-13 is"A"at this site. In addition, 65bp, 310bp, 465bp, 495bp and 510bp all have a base happening mutations. The base of Sull 11-3 at 65bp is T, while that of the other species is C; the bases of Hybrid 46-8, Sull 11-3 and Sull 11-4 at 310bp are all"T", but those of the other species are all C; the base of Sull 11-7 at 465bp is T; the base of Sull 11-3 at 495bp is G, but that of the others is T; the bases of Sull 11-5, 11-13 at 510bp are T, while those of the other races are C. Only two bases happened mutations in the exons, but the mutations have no impact on the amino acid sequences, so it belongs to no righteous mutations. As to the introns, there is no bases mutations happened.3.There was apparent diference of PsCon1 relative quantity in eight races.The PsCon1 relative quantity of Sull 11-3 is highest, but that of Hybrid46-8 is lowest inoculated by strip rust after 12hr. The PsCon1 relative quantity of Sull 11-3 is highest, but that of Sull 11-3 is lowest inoculated by strip rust after 24hr.
