This article through establishes in Longzhong Loess Highlands a half year dry area long time localization experiment, has studied the different cultivation way 8 years later to topsoil soil fungi population's influence.On basis of the principles of soil microbiology and molecularbiology an experiment was conducted to compare the effects of conventional tillage (T) and three conservation tillage practices on population of soil fungi of pea field. The three conservational tillage practices included conventional tillage with straw incorporated (TS), no-till with no straw cover (NT), no-till with stubble retention (NTS). Three conservation tillage practices on pea to soil microbial DNA three different methods of comparison and the type and distribution of microbial populations, the results show that:1. The research of DNA Extraction Methods for Soil fungi⑴Three methods of DNA extraction from Soil fungi all can get to the fragment 23130 bp of DNA, SDS extraction method(method A)DNA is more yield but lowest purity ,SDS-CTAB extraction method(method B) obtained higher purity but the yield is the lowest. Extraction method of combination SDS-CTAB with fungi (method C) obtained the most yield and purity was relatively higher.⑵DNA extraction method of C obtained the most yield,an average of 2.6 times than method A and 10 times than method B.But method A and C obtained lower DNA purity than method B, both of them need to recycle for amplification. After recovered purity of the two methods has improved.But the production after recovered of method C is 1.5 times higher than method A.⑶The DNA through three ways of extraction for PCR amplification.Method A and C after purification and method B all can amplify 250bp fragment.But the method of C band more clear than method B and A ,the effect of amplification is relatively good.2. The research of the effects of different tillage methods on soil fungal population⑴Altogether analyzes eight fungi populations after the sequence, uncultured fungi population has five kinds, three kinds can culture.⑵Each processing soil fungi quantity's sorting is NTS>NT>TS>T. According to different treatment conditions, soil fungi 18SrDNA sequence of AU-PAGE bands, we can see that there are 8 bands NTS processing, NT 6 with handling, TS treatment of 5 bands, T handle 3 bands Which summarizes the NTS> NT> TS> T, the bands represent the number of population number, that means that when the straw till the number of microbial species, followed by a no-tillage without straw, and then followed Conventional tillage straw, and finally the traditional farming.⑶In four kind of processing NTS is most advantageous to edaphon's growth, the soil fungi community's population integer overall presents the protection cultivation to be higher than the traditional cultivation, the straw stalk cover is higher than does not cover.⑷Compared with other similar research literature, the traditional means of molecular biology - urea denaturing gel electrophoresis (AU-PAGE) at the species level is the isolation and identification of new molecular biology methods and findings are not much difference. And it costs much less than other molecular biology methods, simple operation, high success rate.
