The tiger frog (Hoplobatrachus Rugulosus) is widely distributed in the Southern part of China. It is highly appreciated by people because of its unique pleasant aroma, delicious taste and high nutritional value. Because the habitat was destructing and over hunting, the population was decreasing in these years. Even, it disappeared in somewhere. So this species has been listed in AppendixⅡof CITES and as one of the Class II national protected species in China. Also this species has been classified as Vulnerable on IUCN Red List and Species Red List of China. As an important distributed area, Zhejiang province also has experienced drastic decline at population size in recent years. So I studied the genetic diversity of tiger frog in Zhejiang province by the control region of mitochondrial DNA and hoped to provide this species in Zhejiang province.The results are below:1.The content of A,T,G,C are 30.4%-31.9%(average 31.1%),28.9%-30.7%(average 30.0 %),27.1%-28.3%(average 27.7%),10.5%-11.4%(average 11.2%),the AT is reboundance;2. The size is 332 bp,40 variant sites and 282 reserved sites were found. There are 6 singleton variable sites and 34 parsimony informative sites in the variant sites;3. There are 38 haplotypes, the haplotype diversity is 0.983 and the nuceleotide diversity is 0.02226. So the genetic diversity of tiger frog in Zhejiang is high; 4. By the Tajima's test, Tajima's D is -0.36672 (P>0.10), Fs is -18.364 (P>0.10). The result of Tajima's test of selective neutrality was not significant. It revealed that all sequences of Hoplobatrachus Rugulosus were selective neutrality and the population of the Hoplobatrachus Rugulosus in these areas is steady;5. The result reflected by values of K,Dxy and Da is little difference;6. To calculate the genetic distance by MEGA 4.1 and genetic divergence indexes by DNASP4.10 between populations of Hoplobatrachus Rugulosus, the genetic relationship of the ChangShan population and TangXi population, the ChangHua population and the LinAn population is nearly;7. Studying by UPMGA, ME and MP, the result is difference of the SSR. Based on the results above and status of tiger frog farming, I proposed some measures:1. To protect the habitat and adopt a plan of In site conservation or Ex site conservation;2. To maintain the genetic specialty and genetic diversity of breeds, strains and types by combining pedigree file and phyenotypes in the selection of breeders;3. To set up breeding bases of all breeds, strains and types and reasonable genetic management system for preserving populations possessing typical genetic specialty and high genetic diversity and for providing excellent breeders to tiger frog farms.
