Studies On The Method Of Overcoming Fertilization Former Barriers Of Lily And The Relationship Between Lily Anther Chloroplast And Fertility

Several aspects research on Lilium Siberia and White Heaven:1. Studies on the test method of lily pollen viability; 2. Effect of different storage condition on the germination of lily pollen;3. Studies on the method of overcoming fertilization former barriers;4. The impact of light to lily anther ultrastructure.The results are as follows:1. Different components have different effect on lily pollen germination.The sucrose concertration is harmful to pollen germination when it is higher or lover. H3BO3 and CaCl2 can promote pollen germinate;The effect of different culture components is significant with different concertration; The proper culture for pollen germination is sucrose 100g·L-1 +H3BO3 20mg·L-1+ CaCl2 10mg·L-1.2. Used six storage ways,the pollen vigor and germination was tested,we found that the 0 level pollen is longer significantly than 1 level and 2 level;The best and the poor storage way is 0-4℃dry refrigeration and normal storage respectively.The sequence from the best to the poor is 0-4℃cold storage> 0-4℃cold and dry storage >-18-0℃freezing storage>-18-0℃freezing and dry storage>normal tempreture and dry storage> normal tempreture storage.3. Five ways were used to overcome self-hybridization and interspecific hybridization obstacles of lily.The results show that:No matter what is self-hybridization or interspecific hybridization,advance pollination is better than others,and have certain embryo seeds. Cut styles pollination and exogenous hormone promoted ovary swollen,but the capsule falled off early. Exogenous hormone not promote fertilization but have stimulation on ovary swollen. keep style about 0.5cm had better effect on fertilization.4. Using the shading and not shading treatment,anther ultrastructural changes of Oriental lily "Lilium Siberia" was tested. The results show that:Chloroplasts exist in endothecium and middle layer cell of lily anther,and produce starch grains;but there are no starch grains in tapetum; Compared with in unlight,chloroplast retarded and Photosynthesis abate in light of lily(to starch grain size and quantity for standard). During lily anther growth process,chloroplasts produce starch grains continually with photosynthesis of unlight and there are clearly slice layer structure,and disintergration gradually with pollen mature;but chloroplasts produce less and small starch grains, and found slice layer in early detection became fuzzy in light. Anther tapetum will be Programmed cell death in a certain period,but compared with light conditiong,it dissolved early in unlight.
