Silkworm metamorphosis and development by artificial is regarded as one of the fundamental issues in silk industry science.To extend or suspend the pupae pupal stage through the development in optimizing the silk industrial structure has great significance.It's also has great significance in reform the current status of Harvesting and Drying cocoon and achieve major reform by raw silk processing technology.In order to delay the development of silkworm pupae by the dual system GAL4/UAS, we have constructed the activation system vector pigA3GFP-ie-neo-BmWCP4-gal4-fib-polyA and the effect system vector pigA3GFP-ie-neo-UAS -EGT based on piggBac transposon .The activation system vector containing GAL4 gene which is control by the pupal stage specific promoter WCP4 and neomycin resistance gene (neo) which is control by baculovirus ie-1 promoter.The effect system vector is contained ecdysteroid UDP-glucosyltransferase gene (egt) which is control by UAS and neo gene which is control by ie-1 promoter.Through sperm mediated gene transfer(SMGT),we introduced activation system vector and effect system vector into silkworm.By GFP and G418 selection, combined with identification of molecular biology, we preliminary obtained green fluorescence of the silkworm.Initially, we observed that there are no significant difference between the G0 generation of the activation system and the effect system silkworm with the normal silkworm .The result laid the foundation for the pupal stage specific express egt gene in F1 generation.It is also laid the foundation for extending or suspending the pupae pupal stage through the development and Obtaining silkworm varieties material suitable for reeling raw cocoon .In addition, the Wujiang City and Rudong County were chose for investigating the basic situation of sericulture and silkworm disease epidemic. The impact factors on sericulture production in Jiangsu Province was analyzed and summarized based on the survey results. The results showed that the average age of sericulture farmers were 61.9 years and 56.2 years old, the average times undertaking sericultural work reached 27.8 and 26.5 years in Wujiang City and Rudong County, respectively; the sericulture farmers were older. 2 case of silkworm seeds on average were raised for a family every breeding season. the breeding scale for a family was smaller; The cocoons yield for a case of silkworm seeds and 667m2 mulberry gardens were 38.9 kg and 39.2kg, respectively in the spring. the incidence of a disease was 1.272% in spring.The higer loss rate of silkworm is an important factor for the cocoons yield of each case silkworm seeds during the process of raise silkworms. In Wujiang city ,the cocoons yield for a case of silkworm seeds and 667m2 mulberry gardens( are equal in 1 chinese acre) were 65%and42% for Rudong County respectively.The relative concentration of sericulture area in Jiangsu Province is more evident.Compared with 2008, the amount of spring silkworm breeding on average minus 37.29%, the amount of close cocoon minus 32.03%, The reduction of spring cocoons output is by up to 32.60% in every major city of Jiangsu Province in 2009.The relative concentration of sericulture area in Jiangsu Province is more evident.Think regionalization, scale, centralization, diversity is the developing direction of maintains production in jiangsu province.
