Silkworm Bombyx mori(B.mori) colored cocoon varieties are special resources,whose cocoon color controlled by genes.Silkworm colored cocoon varieties can mainly be classified into two families:yellow-red cocoon series that include yellow,pink,golden-yellow and flesh cocoons,and green cocoon series.Pigments in them are absorbed from its host plant,mulberry tree(Morus alba) and from the diet then subsequently modified within the silkworm.At home and abroad,there are many scientific studies on silkworm colored cocoon races about breeding, cocoon characteristics, types and formation mechanisms of pigment, and improving overall economic efficiency and so on,which have come to many valuable theories and practical applied research results.However, it should be given scientific explanations about the metabolism mechanism of pigment,the impact of kinds of pigment and the explicit-implicit relationship of cocoon.By the transfer of pigment in the silkworm research,we can have a further understanding for genetics and mechanism of pigment .It has important academic significance for riching the genetics theory and breeding colored cocoon.We added pigment which was fluorescence into the food of silkworm. We explored the effect of adding pigment. We also used the silkworm variety Dazao as material. To analyze the fluorescent pigment of mulberry, the blood of silkworm, silk gland, cocoon shell, silkworm excrement, silkworm urine and moth urine by TLC.The results obtained are as follows:1,DaZao as the material, the same blue-violet fluorescent pigment which the Rf is 0.35 was obtained from the extract of green cocoon shell, silk gland, silkworm urine, moth urine and mulberry after TLC.The yellow-green fluorescence exist in blood,silk gland and cocoon shell,but it couldn't found in mulberry,silkworm excrement, silkworm urine and moth urine.2,It had little effect on Physiological.The blood,silk gland,silkworm eggs,cocoon shell were colored,and the color was the same with the pigment which adding into the food of silkworm.3,However male or female,the fluorescent pigment were the same.The fluorescent color of the cocoon,blood and silk gland were basically the same.The fluorescent color of the silkworm excrement and urine were the same. But the two fluorescent colors were different.4,The red and yellow fluorescence were mainly in the silkworm blood,silk gland and cocoon shell.Most of the green fluorescence was eliminated from the body of silkworm.5,It was no differences between adding pigment and unadding pigment of silkworm blood protein.Pigment that transport to the silk gland didn't bind protein.6,After adding pigment,we got the red cocoon.We found the pigment of red cocoon in silk fibroin.
