Superoxide dismutase (EC,1.15.1.1) is a metalloenzyme which is widely present in animals, plants and microorganisms. SOD can clear away superoxide anion radical (O2·-) specifically, and also can prevent oxygen free radicals from damaging the cell composition, structure and function, thereby protecting cells from oxidative damage. L-SOD which is rich in content and variety in plants is similar to a class of biologically active natural compounds.Silphium perfoliatum is a former pasture growth in the United States in 1979, introduced from North Korea by the Beijing Botanical Garden, Chinese Academy of Agricultural Sciences to try to grow a successful, after the country's 20 provinces, cities and districts begin to try to grow. After the introduction of domestic Silphium perfoliatum, primary use is as desert plants, sand-fixing, most studies have focused for Silphium perfoliatum in its cultivation, nutritional value based on Silphium perfoliatum, in recent years, the series leaves other applications of loose herbs are also studied, more development and application as forage for their feed palatability and other studies. But, in fact, Silphium perfoliatum has a very rich leaf protein, and for its research in this area and development of domestic rarely reported, so in order for Silphium perfoliatum SOD and SOD-like development, we launched a string of red pine on vanilla in the SOD-conducted applied research in order to make some contribution to their application development.Using single-factor experiment method, with water as solvent extraction Silphium perfoliatum in SOD and SOD-like compounds in the condition that in pH 7.0, solid-liquid ratio 1:7.5, extraction temperature 30℃water bath, under the conditions of extraction 12h, Silphium perfoliatum water extract of the highest activity, up to 31.05U/ml, converted to dry the active material is 232.91U/g.Water extract of Silphium perfoliatum a SOD-simultaneous determination of active substances. Standard curve method, were selected flavonoids, polyphenols and vitamin C three to determine the content, according to the experimental determination of three types of substances in water extracts of Silphium perfoliatum were flavonoid content of 7.9mg/g, polyphenolic compounds 3.84mg/g, Vitamin C 1.26mg/g. Meanwhile, put the liquid antioxidant capacity to take a preliminary determination, select its hydroxyl radical籓H and DPPH radical scavenging capacity measured by ascorbic acid and rutin as a positive control and found that water extract of Silphium perfoliatum of the hydroxyl radical·OH have strong scavenging ability, and lower than the same concentration of ascorbic acid and rutin higher clearance rate, and the water extract of DPPH·free radical scavenging ability also has a strong, results have been very clear with ascorbic acid.Use of Silphium perfoliatum residue after water extraction, ethanol as solvent, its extraction of flavonoids. Experiments using ultrasonic assisted extraction method, using a single experimental factors in the control of flavonoids extracted the best experimental conditions were:90% ethanol concentration, the solid liquid ratio control in 1:15, ultrasonic power of 80%, temperature of ultrasonic extraction 60℃, the role of ultrasound time of 1.5 hours. To the above conditions for the extraction of flavonoids, the maximum extraction rate available to 7.48mg/g of extract.By macroporous resin adsorption and desorption rates, and dynamic adsorption experiments, we macroporous resin Hz 816 and Hz 818 two resins were screened, Hz 818 resin is more suitable for Silphium perfoliatum purified flavonoid extract and use silica gel thin-layer chromatography plate of flavonoids found in Silphium perfoliatum extract contains six major components. After the Hz 818 resin as the medium of flavonoids for the preliminary separation of the solution was found after the sample, the first double deionized water for elution, it can be eluted flavonoids 1, in order to double the volume of 60% ethanol solution can be eluted flavonoids 2,4,5 and 6 and flavonoids depend on macroporous resin ethanol gradient elution separation can not be good, need to be separated with other separation methods.
